Abstract: 本論文分為二部份,第一部份為利用X光繞射法來決定一個金屬配位錯 合物之單晶結構,第二部份為一蛇毒蛋白之養晶方法與過程 第一部份: 晶體為Di( -acetato)bis(dicarboryl-prrazol)ruthenium(I) 此晶體由成大化學系許拱北教授所提供,利用X光繞射來決定其單晶結構, 儀器操作部份由成大地科系董淑芳小姐操作,以Ru原子為中心,屬於六配 位,其幾何結構為扭曲雙金字塔形 第二部份:所養之蛋白質晶體為 Cardiotoxin IV 此蛋白質晶體由清大生命科學系所提供,主 要是利用蒸氣擴散法中的懸滴法來養晶,配合文獻中的50種條件來製造適 合養晶的環境,在這50種條件中再篩選出最適合蛋白質Cardiotoxin IV的 養晶條件 This paper include two parts. One is use X-ray diffraction to determine the structure of a single crystal. The other is the method and process of a crystal growth of protein. First:The single crystal is Di( -acetato)bis(di-carborylprrazol)ruthenium( I) This crystal is from Dr. Shu of NCKU. Use X-ray diffraction to determinethe structure of this single crystal. The Ru atom is the centrum of this structure,it is coordinationnumber 6 and its geometric is distort double pyramid. Second:The crystal of protein is Cardiotoxin IV This crystal is from NTHU. Use hanging drop of vapor diffusion to grow up the crystal of protein. From the paper we can know 50 conditions to grow up the crystal of protein. After experiment we can select the best condition to grow up Cardiotoxin IV between these 50 conditions.
