Item type:Item, A NOVEL ELECTROMAGNETIC TARGETING SYSTEM USING ROTATING MAGNETIC-FLUX CONCENTRATION METHOD FOR NAVIGATING ENDOBRONCHOSCOPY(2018-01-01) ;Chen, Chin-Chung ;Lin, Ching-Kai ;Cheng, Yun-Chien ;Chang, Chen-WeiTsai, Sung-LinIn this paper, we proposed a novel electromagnetic targeting system using rotating magnetic-flux concentration method for navigating endo-bronchoscope. This system consists of a magnetic-flux emitting coil, a magnetic-flux receiving electromagnets-array, a 2D model of bronchial tree, the magnetic-flux concentrator embedded on brush-guiding tube which was connected to the guide sheath. When the concentrator in the 2D bronchial tree passes through the air gap between the emitting coil and the receiving electromagnets array, the concentrator concentrates the magnetic flux between the coil and the array. The concentrated magnetic flux is subsequently received by the receiving electromagnets-array and thus stable voltage output is produced. Furthermore, when the concentrator is rotated, the concentration of the magnetic flux is periodically changed and thereby the voltage output is periodically changed. By analyzing the voltage changes, the location of the concentrator (as well as guide tube and sheath) is targeted. According to the experimental results, the system successfully targets the location of the guide sheath in the 2D model of bronchial tree.Item type:Item, Aerosol-Assisted Plasma Deposition of Biocomposite Coatings: Investigation of Processing Conditions on Coating Properties(2016-12) ;Hsiao, Chun-Ping ;Wu, Cheng-Chen ;Liu, Yung-Hsin ;Yang, Yi-WeiCheng, Yun-ChienThe aerosol-assisted atmospheric-pressure planar dielectric-barrier-discharge-type plasma deposition (AA-APPD) system can polymerize monomers and immobilize biomolecules simultaneously. In this paper, we used this technology to deposit biocomposite thin films of polyethylene (PE) along with lysozyme (Lyz) under different aerosol solution temperatures and at different coating positions in the discharge region. In addition, we compared the deposition efficacy of two kinds of Lyz solution using deionized (DI)-water and phosphate-buffered saline (PBS) as solvents, respectively. The results show that deposition rate of thin films increases with increasing solution temperature at most coating positions. In addition, coatings from Lyz-PBS perform higher deposition rates, but with weaker Lyz viability as compared with that of Lyz DI-water. Finally, we also verified that this DBD-type AA-APPD system can not only successfully deposit Lyz in PE thin film, but also maintain Lyz viability after plasma process under different solution temperatures and positions.Item type:Item, Aerosol-Assisted Plasma Deposition of Biocomposite Coatings: Investigation of Processing Conditions on Coating Properties(2016-12-01) ;Hsiao, Chun-Ping ;Wu, Cheng-Chen ;Liu, Yung-Hsin ;Yang, Yi-WeiCheng, Yun-ChienThe aerosol-assisted atmospheric-pressure planar dielectric-barrier-discharge-type plasma deposition (AA-APPD) system can polymerize monomers and immobilize biomolecules simultaneously. This system consists of a magnetic-flux emitting electromagnets-arrays fixed on a semi-circular mechanicalsupport, magnetic-flux receiving electromagnets, and a magnetic-flux concentrator (i.e., silicon-steel collar) which fixed on distal end of a guide sheath of the endo-bronchoscope. In initial state, when the emitting electromagnets produce magnetic flux in sequence, the receiving electromagnets receive the magnetic flux and consequently produce voltage outputs by the electromagnetic induction. When the silicon-steel collar with the guide sheath travels through the system, the magnetic flux is concentrated by the collar. Thus, the voltage outputs of the receiving electromagnets are changed. By analyzing the change ofvoltage outputs, the location ofthe silicon-steel collar with the guide sheath is obtained/targeted. This means that the location of the endo-bronchoscope is also targeted.Item type:Item, Analysis of Hydroxyl Radical and Hydrogen Peroxide Generated in Helium-Based Atmospheric-Pressure Plasma Jet and in Different Solutions Treated by Plasma for Bioapplications(2020-06-22) ;Chen, Tsung-Wen ;Liu, Chih-Tung ;Chen, Chao-Yu ;Wu, Mu-ChienChien, Po-ChienThis study systematically analyzed reactive species generated with self-built helium-based low-temperature atmospheric-pressure plasma jet (He-APPJ); we measured the hydroxyl radical (OH) and hydrogen peroxide (H2O2) above surfaces of plasma-treating solutions and in plasma-treated medium (PTM) of different solutions, including deionized water, phosphate buffered saline, and Dulbecco's modified Eagle medium. The effects of adding O(2)into He working gas on OH and H(2)O(2)generation were discussed. The MCF7 cell responses to plasma treatment were also observed. The OH emission (309 nm) in He-APPJ and above surfaces of plasma-treating solutions were analyzed through optical emission spectroscopy, and concentrations of OH and H(2)O(2)in different solutions were measured using terephthalic acid and Amplex Red, respectively. In our plasma system, it was observed that higher conductivity of solutions may cause stronger OH emission above solution. The ingredients of the solution may affect the concentrations of OH and H2O2. Addition of 0.1% O(2)generates strongest OH emission above surfaces of plasma-treating solution with our He-APPJ, and thus, the highest H(2)O(2)concentration in PTM. The MCF7 cell viability is influenced by the H(2)O(2)concentration in PTM. The results of this study can facilitate further study of plasma effects on PTM and cell culture.Item type:Item, Enhancing adhesion and polymerization of lipase-plasma-polymerized-ethylene coatings deposited with planar dielectric-barrier-discharge-type aerosol-assisted atmospheric-pressure plasma system(2018-04-01) ;Cheng, Yun-Chien ;Hsiao, Chun-Ping ;Liu, Yung-Hsin ;Yang, Chu-HaoChiang, Chun-YiTo enhance the adhesion and polymerization of lipase-plasma-polymerized-ethylene (lipase-PPE) coatings deposited by aerosol-assisted atmospheric plasma deposition (AAAPPD), the effects of voltage, ceramic-dielectric thickness, and pre-coated PPE layer were investigated. In AAAPPD system, ethylene and lipase-phosphate-buffered-saline aerosol were used as precursors to polymerize ethylene and embed lipase simultaneously. The deposited lipase-PPE coatings were analyzed with FTIR, SEM, and surface profiler. The results show that increasing voltage from 3 to 4 kV and decreasing ceramic-dielectric thickness from 1 to 0.38 mm improved the coating adhesion and polymerization. Also, depositing a PPE layer with high energy prior to lipase-PPE coating enhanced adhesion. These improvements of the biomolecule-PPE coatings facilitate further bioapplications of this new biomolecule-embedding method.Item type:Item, Enhancing the Microparticle Deposition Stability and Homogeneity on Planer for Synthesis of Self-Assembly Monolayer(2018-03-01) ;Shih, An-Ci ;Han, Chi-Jui ;Kuo, Tsung-Cheng ;Cheng, Yun-Chien機械工程學系The deposition stability and homogeneity of microparticles improved with mask, lengthened nozzle and flow rate adjustment. The microparticles can be used to encapsulate monomers, before the monomers in the microparticles can be deposited onto a substrate for nanoscale self-assembly. For the uniformity of the synthesized nanofilm, the homogeneity of the deposited microparticles becomes an important issue. Based on the ANSYS simulation results, the effects of secondary flow were minimized with a lengthened nozzle. The ANSYS simulation was also used to investigate the ring-vortex generation and why the ring vortex can be eliminated by adding a mask with an aperture between the nozzle and deposition substrate. The experimental results also showed that particle deposition with a lengthened nozzle was more stable, while adding the mask stabilized deposition and diminished the ring-vortex contamination. The effects of flow rate and pressure were also investigated. Hence, the deposition stability and homogeneity of microparticles was improved.Item type:Item, Passive Magnetic-Flux-Concentrator Based Electromagnetic Targeting System for Endobronchoscopy(2019-12-01) ;Chen, Chin-Chung ;Lin, Ching-Kai ;Chang, Chen-Wei ;Cheng, Yun-ChienChen, Jia-EnIn this paper, we demonstrate an innovative electromagnetic targeting system utilizing a passive magnetic-flux-concentrator for tracking endobronchoscope used in the diagnosis process of lung cancer tumors/lesions. The system consists of a magnetic-flux emitting coil, a magnetic-flux receiving electromagnets-array, and high permeability silicon-steel sheets rolled as a collar (as the passive magnetic-flux-concentrator) fixed in a guide sheath of an endobronchoscope. The emitting coil is used to produce AC magnetic-flux, which is consequently received by the receiving electromagnets-array. Due to the electromagnetic-induction, a voltage is induced in the receiving electromagnets-array. When the endobronchoscope's guide sheath (with the silicon-steel collar) travels between the emitting coil and the receiving electromagnets-arrays, the magnetic flux is concentrated by the silicon-steel collar and thereby the induced voltage is changed. Through analyzing the voltage-pattern change, the location of the silicon-steel collar with the guide sheath is targeted. For testing, a bronchial-tree model for training medical doctors and operators is used to test our system. According to experimental results, the system is successfully verified to be able to target the endobronchoscope in the bronchial-tree model. The targeting errors on the x-, y- and z-axes are 9 mm, 10 mm, and 5 mm, respectively.Item type:Item, Protein Crosslinking and Immobilization in 3D Microfluidics through Multiphoton Absorption(2020-01-12) ;Lin, Chen-Feng ;Su, Che-Fu ;Lin, Kung-Hsuan ;Hsieh, Yu-ShenCheng, Yun-ChienThis study used laser multiphoton absorption (MPA) to crosslink and immobilize protein in 3D microfluidics. MPA can be used to crosslink proteins to form smaller crosslinked-protein structures than those formed through single-photon absorption. However, studies on MPA protein crosslinking in 3D microfluidics are lacking. In this study, the MPA was implemented using pulse laser to induce bovine serum albumin (BSA) crosslinking. We also applied (3-glycidyloxypropyl)trimethoxysilane to increase the adhesion of crosslinked BSA. Also, the width of crosslinked-BSA structure was measured, and then curve-fitting and optical equation were used to predict heights of the crosslinked-BSA structures formed under various laser powers. At last, BSA structures were effectively crosslinked in the three-layered glass or polydimethylsiloxane microchannels. In contrast to studies on single-photon absorption in 3D microfluidics or MPA protein crosslinking in 2D microfluidics, we obtained relatively small crosslinked-BSA structures through MPA and immobilize BSA in 3D microfluidics. In the future, this method can be applied in monolithic 3D microfluidics for protein crosslinking to avoid misalignment and leakage in layer-by-layer 3D microchannels and form more compact 3D microfluidics. These features are conducive to enhance the diversity and convenience of microfluidics in biomedical testing.Item type:Item, Synergistic effects of plasma-activated medium and chemotherapeutic drugs in cancer treatment(2018-04-04) ;Chen, Chao-Yu ;Cheng, Yun-Chien ;Cheng, Yi-Jing ;機械工程學系Department of Mechanical EngineeringChemotherapy is an important treatment method for metastatic cancer, but the drug-uptake efficiency of cancer cells needs to be enhanced in order to diminish the side effects of chemotherapeutic drugs and improve survival. The use of a nonequilibrium low-temperature atmospheric-pressure plasma jet (APPJ) has been demonstrated to exert selective effects in cancer therapy and to be able to enhance the uptake of molecules by cells, which makes an APPJ a good candidate adjuvant in combination chemotherapy. This study estimated the effects of direct helium-based APPJ (He-APPJ) exposure (DE) and He-APPJ-activated RPMI medium (PAM) on cell viability and migration. Both of these treatments decreased cell viability and inhibited cell migration, but to different degrees in different cell types. The use of PAM as a culture medium resulted in the dialkylcarbocyanine (DiI) fluorescent dye entering the cells more efficiently. PAM was combined with the anticancer drug doxorubicin (Doxo) to treat human heptocellular carcinoma HepG2 cells and human adenocarcinomic alveolar basal epithelial A549 cells. The results showed that the synergistic effects of combined PAM and Doxo treatment resulted in stronger lethality in cancer cells than did PAM or Doxo treatment alone. To sum up, PAM has potential as an adjuvant in combination with other drugs to improve curative cancer therapies.Item type:Item, Tooth bleaching by using a helium-based low-temperature atmospheric pressure plasma jet with saline solution(2017-11-01) ;Cheng, Yun-Chien ;Wu, Chia-Hua ;Liu, Chih-Tung ;Lin, Chia-YungChiang, Hsuan-PingIn this study, tooth bleaching was performed by using a helium-based low temperature atmospheric-pressure plasma jet (He-APPJ) with saline solution to avoid ozone and hydrogen peroxide (H2O2) harm. In this study, tooth samples were treated through three approaches, namely a helium flow with saline, H2O2 gel, and He-APPJ with saline. The bleaching efficacy of the He-APPJ treatment was the highest among the three groups. Furthermore, scanning electron microscopy images showed that enamel treated with He-APPJ was much less damaged than that treated with H2O2 gel. In addition, ozone tests showed that APPJ produced little ozone (<0.35ppm). In conclusion, He-APPJ with saline solution achieves higher bleaching efficacy and is less harmful than methods that use H2O2 gel.Item type:Item, Using aerosol-assisted atmospheric-pressure plasma to embed proteins onto a substrate in one step for biosensor fabrication(2018-09-01) ;Liu, Yung-Hsin ;Yang, Chu-Hao ;Lin, Tsung-Rong ;Cheng, Yun-Chien機械工程學系This study shows that aerosol-assisted dielectric-barrier-discharge atmospheric-pressure plasma deposition (AAAPPD) is a rapid and reagent-free method to entrap proteins on different substrates for biosensor fabrication, and the proteins were not damaged. AAAPPD involves depositing plasma-polymerized ethylene (ppE) with grafted hydroxyl functional groups and embedding the protein in the ppE in one step, making the protein entrapment faster than conventional methods and without using reagents. In this study, SEM and XPS show that more protein deposited near the inlet. Electrophoresis, temperature analysis, and immunostaining respectively demonstrated that the protein maintained its integrity, was unheated, and exhibited activity during AAAPPD. The immunostaining results of AAAPPD protein was close to that of covalent-bonded protein. AAAPPD also entraps protein on three different biosensor substrates.Item type:Item, Using Micro-Molding and Stamping to Fabricate Conductive Polydimethylsiloxane-Based Flexible High-Sensitivity Strain Gauges(2018-02-01) ;Han, Chi-Jui ;Chiang, Hsuan-Ping ;Cheng, Yun-Chien ;機械工程學系Department of Mechanical EngineeringIn this study, polydimethylsiloxane (PDMS) and conductive carbon nanoparticles were combined to fabricate a conductive elastomer PDMS (CPDMS). A high sensitive and flexible CPDMS strain sensor is fabricated by using stamping-process based micro patterning. Compared with conventional sensors, flexible strain sensors are more suitable for medical applications but are usually fabricated by photolithography, which suffers from a large number of steps and difficult mass production. Hence, we fabricated flexible strain sensors using a stamping-process with fewer processes than photolithography. The piezoresistive coefficient and sensitivity of the flexible strain sensor were improved by sensor pattern design and thickness change. Micro-patterning is used to fabricate various CPDMS microstructure patterns. The effect of gauge pattern was evaluated with ANSYS simulations. The piezoresistance of the strain gauges was measured and the gauge factor determined. Experimental results show that the piezoresistive coefficient of CPDMS is approximately linear. Gauge factor measurement results show that the gauge factor of a 140.0 mu m thick strain gauge with five grids is the highest.
