Item type:Item, Automatic and objective oral cancer diagnosis by Raman spectroscopic detection of keratin with multivariate curve resolution analysis(2016-01-25) ;Chen, Po-Hsiung ;Shimada, Rintaro ;Yabumoto, Sohshi ;Okajima, HajimeAndo, MasahiroWe have developed an automatic and objective method for detecting human oral squamous cell carcinoma (OSCC) tissues with Raman microspectroscopy. We measure 196 independent Raman spectra from 196 different points of one oral tissue sample and globally analyze these spectra using a Multivariate Curve Resolution (MCR) analysis. Discrimination of OSCC tissues is automatically and objectively made by spectral matching comparison of the MCR decomposed Raman spectra and the standard Raman spectrum of keratin, a well-established molecular marker of OSCC. We use a total of 24 tissue samples, 10 OSCC and 10 normal tissues from the same 10 patients, 3 OSCC and 1 normal tissues from different patients. Following the newly developed protocol presented here, we have been able to detect OSCC tissues with 77 to 92% sensitivity (depending on how to define positivity) and 100% specificity. The present approach lends itself to a reliable clinical diagnosis of OSCC substantiated by the "molecular fingerprint" of keratin.Item type:Item, Communication: Three-dimensional model for phonon confinement in small particles: Quantitative bandshape analysis of size-dependent Raman spectra of nanodiamonds(2014-01-28) ;Korepanov, Vitaly I. ;Witek, Henryk ;Okajima, Hajime ;Osawa, EijiHamaguchi, Hiro-ORaman spectroscopy of nano-scale materials is facing a challenge of developing a physically sound quantitative approach for the phonon confinement effect, which profoundly affects the phonon Raman band shapes of small particles. We have developed a new approach based on 3-dimensional phonon dispersion functions. It analyzes the Raman band shapes quantitatively in terms of the particle size distributions. To test the model, we have successfully obtained good fits of the observed phonon Raman spectra of diamond nanoparticles in the size range from 1 to 100 nm. In the present work, we try to verify the efficacy of using the polymorphic features of fats for discriminating animal-fat origins. We use Raman spectroscopy to collect the structural information of fat crystals. It is shown that a single Raman band at 1417 cm(-1) successfully differentiates pork fats from beef fats. This band is known to be characteristic of the beta'-polymorph of fats. Pork fats show this band because they contain the beta'-polymorph after rapid cooling to 0 degrees C. In beef pork-fat mixtures, this hand is not detected even in the presence of 50% pork fat; an addition of beef fat to pork fat is likely to produce a mixed fat with a completely different polymorphic behavior. This method seems to have the potential to detect beef products contaminated with pork-adipose tissue.Item type:Item, Local structure formation in alkyl-imidazolium-based ionic liquids as revealed by linear and nonlinear Raman spectroscopy(2007-11-01) ;Iwata, Koichi ;Okajima, Hajime ;Saha, Satyen ;Hamaguchi, Hiro-O應用化學系We show several pieces of Raman spectroscopic evidence that are indicative of local structure formation in imidazolium-based ionic liquids. Low-frequency Raman spectra of C(n)mimX, where C(n)mim stands for 1-alkyl(CnH2n+1)-3-methylimidazolium cation and X represents the anion, exhibit broad bands assignable to collective modes of local structures. Spatial distributions of coherent anti-Stokes Raman scattering (CARS) signals from C(n)mim[PF(6)l are consistent with local structures whose size increases with increasing n. Picosecond Raman spectra of S, trans-stilbene as a "picosecond Raman thermometer" show microscopic thermal inhomogeneity ascribable to local structure formation in C(2)mimTf(2)N and C(4)mimTf(2)N. We also describe two novel phenomena that we believe are relevant to extraordinary nanoenvironments generated by local structures in a magnetic ionic liquid C(4)mim[FeCl4].Item type:Item, Photobleaching of the "Raman Spectroscopic Signature of Life" and Mitochondrial Activity in Rho(-) Budding Yeast Cells(2009-08-06) ;Onogi, Chikao ;Hamaguchi, Hiro-O ;應用化學系 ;應用化學系分子科學碩博班Department of Applied ChemistryRaman spectra of mitochondria in single living budding yeast (zygote of Saccharomyces cerevisiae and Saccharomyces bayanus) cells have been measured and compared for two types of tetraploid strains, the wild-type strain and the rho(-) strain. The "Raman spectroscopic signature of life", a strong Raman band at 1602 cm(-1) that sharply reflects the mitochondrial activity, has been found in both of the two strains with similar intensities. This signature showed photobleaching under irradiation of the 632.8 nm line of a He-Ne laser, only for the rho(-) strain. The photobleaching recovered by keeping the cell in the dark for two hours without laser irradiation. These findings indicate that the molecular species that gives rise to the 1602 cm(-1) band is an intermediate formed and accumulated during the metabolic cycle and that it absorbs at 632.8 nm to undergo photodecomposition. We also found a number of weak Raman bands in the 400-1200 cm(-1) region that show the same temporal behaviors as that of the 1602 cm(-1) band by an SVD analysis of time-resolved Raman spectra, Several new pieces of information oil the origin of the "Raman spectroscopic signature of life" have thus been obtained.
