Item type:Item, 2xVDD-Tolerant Power-Rail ESD Clamp Circuit With Low Standby Leakage in 65-nm CMOS Process(2010) ;Lin, Chun-Yu ;Ker, Ming-Dou ;電機學院College of Electrical and Computer EngineeringWith the consideration of low standby leakage in nanoscale CMOS processes, a new 2xVDD-tolerant ESD clamp circuit was presented in this paper. The new ESD clamp circuit had a high-voltage-tolerant ESD detection circuit to improve the turn-on efficiency of the silicon-controlled-rectifier-based (SCR-based) ESD device. This design had been successfully verified in a 65-nm CMOS process. The leakage current of this ESD clamp circuit under normal circuit operating condition was only similar to 200 nA. Besides, this ESD clamp circuit can achieve 4.8-kV HBM ESD robustness. Therefore, this design was very suitable for mixed-voltage I/O interfaces in nanoscale CMOS processes.Item type:Item, 3D surface morphology imaging of opaque microstructures via light-field microscopy(2018-07-12) ;Da Sie, Yong ;Lin, Chun-Yu ;Chen, Shean-Jen ;光電學院College of PhotonicsObserving dynamic micro-scale phenomena occurring at millisecond time scales, such as organism activity, micron particle flows, or any opaque object observation, requires volumetric microscopy techniques able to achieve high data acquisition rates while maintaining contrast so that measurement of fine micro-scale features is possible. In realizing this purpose, the light-field (LF) technique has already been used on three-dimensional (3D) scene capturing and even for microscopic visualizations. In studying the ability and feasibility of 3D surface morphology reconstruction via LF microscopy, we adopted a lab-made LF microscope and integrated a four-dimensional Fourier slice algorithm and a Markov random field propagation algorithm. Furthermore, for numerical comparison and quantized analysis, the Tenengrad function was utilized to calculate the average contrast of the region of interest. Reflective US Air Force targets and 3D photolithography-made micro-scaffolds coated with 50 nm nickel thin films were adopted for system alignment and calibration. The experimental results demonstrate that the developed LF microscope with the signal processing algorithms can observe the 3D surface morphology of opaque microstructures with one snapshot, and has been preliminary applied to Brownian motion observation with 30 Hz volumetric image rate.Item type:Item, A Fully Integrated 8-Channel Closed-Loop Neural-Prosthetic CMOS SoC for Real-Time Epileptic Seizure Control(2014-01-01) ;Chen, Wei-Ming ;Chiueh, Herming ;Chen, Tsan-Jieh ;Ho, Chia-LunJeng, ChiAn 8-channel closed-loop neural-prosthetic SoC is presented for real-time intracranial EEG (iEEG) acquisition, seizure detection, and electrical stimulation in order to suppress epileptic seizures. The SoC is composed of eight energy-efficient analog front-end amplifiers (AFEAs), a 10-b delta-modulated SAR ADC (DMSAR ADC), a configurable bio-signal processor (BSP), and an adaptive high-voltage-tolerant stimulator. A wire-less power-and-data transmission system is also embedded. By leveraging T-connected pseudo-resistors, the high-pass (low-pass) cutoff frequency of the AFEAs can be adjusted from 0.1 to 10 Hz (0.8 to 7 kHz). The noise-efficiency factor (NEF) of the AFEA is 1.77, and the DMSAR ADC achieves an ENOB of 9.57 bits. The BSP extracts the epileptic features from time-domain entropy and frequency spectrum for seizure detection. A constant 30-mu A stimulus current is delivered by closed-loop control. The acquired signals are transmitted with on-off keying (OOK) modulation at 4 Mbps over the MedRadio band for monitoring. A multi-LDO topology is adopted to mitigate the interferences across different power domains. The proposed SoC is fabricated in 0.18-mu m CMOS and occupies 13.47 mm(2). Verified on Long Evans rats, the proposed SoC dissipates 2.8 mW and achieves high detection accuracy 92% within 0.8 s.Item type:Item, A Fully Integrated 8-Channel Closed-Loop Neural-Prosthetic SoC for Real-Time Epileptic Seizure Control(2013-01-01) ;Chen, Wei-Ming ;Chiueh, Herming ;Chen, Tsan-Jieh ;Ho, Chia-LunJeng, ChiItem type:Item, A High-Voltage-Tolerant Stimulator Realized in the Low-Voltage CMOS Process for Cochlear Implant(2014-01-01) ;Lin, Kuan-Yu ;Ker, Ming-Dou ;Lin, Chun-Yu ;生醫電子轉譯研究中心Biomedical Electronics Translational Research CenterA biomedical stimulator with four high-voltage-tolerant output channels, combined with on-chip positive high voltage generator, is proposed. For the purpose of integration with other circuit blocks into a system-on-chip (SoC) for cochlear implant biomedical applications, this design has been realized with the 1.8-V/3.3-V transistors in a 0.18-mu m CMOS process. This stimulator only needs one single supply voltage of 1.8 V, but the maximum stimulation voltage can be as high as 7 V. The dynamic bias technique and stacked MOS configuration are used to implement this stimulator in the low-voltage CMOS process, without causing the issues of electrical overstress and gate-oxide reliability during circuit operation.Item type:Item, A rapid and quantitative method to detect human circulating tumor cells in a preclinical animal model(2017-06-23) ;Tu, Shih-Hsin ;Hsieh, Yi-Chen ;Huang, Li-Chi ;Lin, Chun-YuHsu, Kai-WenBackground: As cancer metastasis is the deadliest aspect of cancer, causing 90% of human deaths, evaluating the molecular mechanisms underlying this process is the major interest to those in the drug development field. Both therapeutic target identification and proof-of-concept experimentation in anti-cancer drug development require appropriate animal models, such as xenograft tumor transplantation in transgenic and knockout mice. In the progression of cancer metastasis, circulating tumor cells (CTCs) are the most critical factor in determining the prognosis of cancer patients. Several studies have demonstrated that measuring CTC-specific markers in a clinical setting (e.g., flow cytometry) can provide a current status of cancer development in patients. However, this useful technique has rarely been applied in the real-time monitoring of CTCs in preclinical animal models. Methods: In this study, we designed a rapid and reliable detection method by combining a bioluminescent in vivo imaging system (IVIS) and quantitative polymerase chain reaction (QPCR)-based analysis to measure CTCs in animal blood. Using the IVIS Spectrum CT System with 3D-imaging on orthotropic-developed breast-tumor-bearing mice. Results: In this manuscript, we established a quick and reliable method for measuring CTCs in a preclinical animal mode. The key to this technique is the use of specific human and mouse GUS primers on DNA/RNA of mouse peripheral blood under an absolute qPCR system. First, the high sensitivity of cancer cell detection on IVIS was presented by measuring the luciferase carried MDA-MB-231 cells from 5 to 5x10(11) cell numbers with great correlation (R-2 = 0.999). Next, the MDA-MB-231 cell numbers injected by tail vein and their IVIS radiance signals were strongly corrected with qPCR-calculated copy numbers (R-2 > 0.99). Furthermore, by applying an orthotropic implantation animal model, we successfully distinguished xenograft tumor-bearing mice and control mice with a significant difference (p < 0.001), whereas IVIS Spectrum-CT 3D-visualization showed that blood of mice with lung metastasis contained more than twice the CTC numbers than ordinary tumor-bearing mice. We demonstrated a positive correlation between lung metastasis status and CTC numbers in peripheral mouse blood. Conclusion: Collectively, the techniques developed for this study resulted in the integration of CTC assessments into preclinical models both in vivo and ex vivo, which will facilitate translational targeted therapy in clinical practice.Item type:Item, A Wireless Human Motion Capturing System for Home Rehabilitation(2009) ;Tseng, Yu-Chee ;Wu, Chin-Hao ;Wu, Fang-Jing ;Huang, Chi-FuKing, Chung-TaFollowing the trend of miniature intelligent sensing, wearing small, integrated wireless sensor nodes, such as one with accelerometers and compasses, to capture human body motions may have many applications in medical care and computer animation. In this paper, we demonstrate the use of intelligent sensors to capture human motions for home rehabilitation. We design a game to help a patient to conduct his/her rehabilitation program. For each exercise, the patient is instructed to wear sensors on specified movable body parts. The system will then estimate the quality of the movements and give scores as if it is advised by a therapist. In this way, patients will no longer feel painful and boring as that in traditional rehabilitation, which is typically done in hospitals.Item type:Item, ABLGenomic Editing Sufficiently Abolishes Oncogenesis of Human Chronic Myeloid Leukemia Cells In Vitro and In Vivo(2020-06-01) ;Chen, Shu-Huey ;Hsieh, Yao-Yu ;Tzeng, Huey-En ;Lin, Chun-YuHsu, Kai-WenChronic myelogenous leukemia (CML) is the most common type of leukemia in adults, and more than 90% of CML patients harbor the abnormal Philadelphia chromosome (Ph) that encodes the BCR-ABL oncoprotein. Although the ABL kinase inhibitor (imatinib) has proven to be very effective in achieving high remission rates and improving prognosis, up to 33% of CML patients still cannot achieve an optimal response. Here, we used CRISPR/Cas9 to specifically target theBCR-ABLjunction region in K562 cells, resulting in the inhibition of cancer cell growth and oncogenesis. Due to the variety ofBCR-ABLjunctions in CML patients, we utilized gene editing of the humanABLgene for clinical applications. Using theABLgene-edited virus in K562 cells, we detected 41.2% indels inABLsgRNA_2-infected cells. TheABL-edited cells reveled significant suppression of BCR-ABL protein expression and downstream signals, inhibiting cell growth and increasing cell apoptosis. Next, we introduced theABLgene-edited virus into a systemic K562 leukemia xenograft mouse model, and bioluminescence imaging of the mice showed a significant reduction in the leukemia cell population inABL-targeted mice, compared to the scramble sgRNA virus-injected mice. In CML cells from clinical samples, infection with theABLgene-edited virus resulted in more than 30.9% indels and significant cancer cell death. Notably, no off-target effects or bone marrow cell suppression was found using theABLgene-edited virus, ensuring both user safety and treatment efficacy. This study demonstrated the critical role of theABLgene in maintaining CML cell survival and tumorigenicity in vitro and in vivo.ABLgene editing-based therapy might provide a potential strategy for imatinib-insensitive or resistant CML patients.Item type:Item, An Integrated Genomic Strategy to Identify CHRNB4 as a Diagnostic/Prognostic Biomarker for Targeted Therapy in Head and Neck Cancer(2020-05-01) ;Chuang, Yi-Hsuan ;Lee, Chia-Hwa ;Lin, Chun-Yu ;Liu, Chia-LinHuang, Sing-HanAlthough many studies have shown the association between smoking and the increased incidence and adverse prognosis of head and neck squamous cell carcinoma (HNSCC), the mechanisms and pharmaceutical targets involved remain unclear. Here, we integrated gene expression signatures, genetic alterations, and survival analyses to identify prognostic indicators and therapeutic targets for smoking HNSCC patients, and we discovered that the FDA-approved drug varenicline inhibits the target for cancer cell migration/invasion. We first identified 18 smoking-related and prognostic genes for HNSCC by using RNA-Seq and clinical follow-up data. One of these genes, CHRNB4 (neuronal acetylcholine receptor subunit beta-4), increased the risk of death by approximately threefold in CHRNB4-high expression smokers compared to CHRNB4-low expression smokers (log rank, p = 0.00042; hazard ratio, 2.82; 95% CI, 1.55-5.14), former smokers, and non-smokers. Furthermore, we examined the functional enrichment of co-regulated genes of CHRNB4 and its 246 frequently occurring copy number alterations (CNAs). We found that these genes were involved in promoting angiogenesis, resisting cell death, and sustaining proliferation, and contributed to much worse outcomes for CHRNB4-high patients. Finally, we performed CHRNB4 gene editing and drug inhibition assays, and the results validate these observations. In summary, our study suggests that CHRNB4 is a prognostic indicator for smoking HNSCC patients and provides a potential new therapeutic drug to prevent recurrence or distant metastasis.Item type:Item, Area-Efficient and Low-Leakage Diode String for On-Chip ESD Protection(2016-02-01) ;Lin, Chun-Yu ;Wu, Po-Han ;Ker, Ming-Dou ;電子工程學系及電子研究所Department of Electronics Engineering and Institute of ElectronicsDiode string was used as the effective on-chip electrostatic discharge (ESD) protection device. To reduce the leakage current and the layout area, an area-efficient and low-leakage diode string is proposed in this paper. The standard steps of P- implantation and silicide blocking in CMOS process are used in this design to realize the proposed diode string with stacked P-/N+ diodes. The test devices of the proposed design have successfully been verified in the silicon chip. With the high ESD robustness, low leakage current, and small layout area, the proposed diode string can be a better solution for on-chip ESD protection applications.Item type:Item, Characteristics of scrub typhus, murine typhus, and Q fever among elderly patients: Prolonged prothrombin time as a predictor for severity(2019-02-01) ;Chang, Ko ;Lee, Nan-Yao ;Ko, Wen-Chien ;Lin, Wei-RuChen, Yen-HsuBackground/purpose: The clinical manifestations of scrub typhus, murine typhus and acute Q fever in the elderly are not clear. Methods: We conducted a retrospective study to identify the characteristics of the elderly aged >= 65 years with a comparison group aged 18-64 years among patients with scrub typhus, murine typhus, or acute Q fever who were serologically confirmed at three hospitals in Taiwan during 2002-2011. Results: Among 441 cases, including 187 cases of scrub typhus, 166 acute Qfever, and 88 murine typhus, 68 (15.4%) cases were elderly patients. The elderly had a higher severe complication rate (10.3% vs. 3.5%, p = 0.022), but did not have a significantly higher mortality rate (1.47% vs. 0.54%, p = 0.396). Compared with those without severe complications, we found the elderly (p = 0.022), dyspnea (p = 0.006), less relative bradycardia (p = 0.004), less febrile illness (p = 0.004), prolonged prothrombin time (PT) (p = 0.002), higher levels of initial C-reactive protein (p = 0.039), blood leukocyte counts (p = 0.01), and lower platelet counts (p = 0.012) are significantly associated with severe complications. Only prolonged prothrombin time was associated with severe complications in multivariate analysis (p = 0.018, CI 95% 0.01-0.66). Among clinical symptoms and laboratory data, multivariate analysis revealed chills was less frequently occurred in the elderly (p = 0.012, 95% confidence interval [CI]: 1.33-9.99). Conclusion: The elderly cases with scrub typhus, murine typhus, or acute Q fever would be more likely to have severe complications, for which prothrombin time prolongation is an important predictor for severe complications. However, the thinner gate oxide and silicided drain/source in nanoscale CMOS technologies seriously degrade the electrostatic discharge (ESD) robustness of RF circuits. Against ESD damage, on-chip ESD protection design must be included in RF circuits. As the RF circuits operating in the higher frequency band, the parasitic effect from ESD protection devices and/or circuits must be strictly limited. To provide the effective ESD protection for a 60-GHz low-noise amplifier (LNA) with less RF performance degradation, a new ESD protection design was studied in a 65-nm CMOS process. Such ESD-protected LNA with simulation/measurement results has been successfully verified in silicon chip to to achieve the 2-kV HBM ESD robustness with the lower power loss in a smaller layout area.Item type:Item, CRISPR/Cas9 Genome Editing of Epidermal Growth Factor Receptor Sufficiently Abolished Oncogenicity in Anaplastic Thyroid Cancer(2018-01-01) ;Huang, Li-Chi ;Tam, Ka-Wai ;Liu, Wei-Ni ;Lin, Chun-YuHsu, Kai-WenAnaplastic carcinoma of the thyroid (ATC), also called undifferentiated thyroid cancer, is the least common but most aggressive and deadly thyroid gland malignancy of all thyroid cancers. The aim of this study is to explore essential biomarker and use CRISPR/Cas9 with lentivirus delivery to establish a gene-target therapeutic platform in ATC cells. At the beginning, the gene expression datasets from 1036 cancers from CCLE and 8215 tumors from TCGA were collected and analyzed, showing EGFR is predominantly overexpressed in thyroid cancers than other type of cancers (P = 0 017 in CCLE and P = 0 001 in TCGA). Using CRISPR/Cas9 genomic edit system, ATC cells with EGFR sgRNA lentivirus transfection obtained great disruptions on gene and protein expression, resulting in cell cycle arrest, cell growth inhibition, and most importantly metastasis turn-off ability. In addition, the FDA-approved TKI of afatinib for EGFR targeting also illustrates great anticancer activity on cancer cell death occurrence, cell growth inhibition, and cell cycle arrest in SW579 cells, an EGFR expressing human ATC cell line. Furthermore, off-target effect of using EGFR sgRNAs was measured and found no genomic editing can be detected in off-target candidate gene. To conclude, this study provides potential ATC therapeutic strategies for current and future clinical needs, which may be possible in increasing the survival rate of ATC patients by translational medicine.Item type:Item, Cyclooxygenase-2 facilitates dengue virus replication and serves as a potential target for developing antiviral agents(2017-03-20) ;Lin, Chun-Kuang ;Tseng, Chin-Kai ;Wu, Yu-Hsuan ;Liaw, Chih-ChuangLin, Chun-YuCyclooxygenase-2 (COX-2) is one of the important mediators of inflammation in response to viral infection, and it contributes to viral replication, for example, cytomegalovirus or hepatitis C virus replication. The role of COX-2 in dengue virus (DENV) replication remains unclear. In the present study, we observed an increased level of COX-2 in patients with dengue fever compared with healthy donors. Consistent with the clinical data, an elevated level of COX-2 expression was also observed in DENV-infected ICR suckling mice. Using cell-based experiments, we revealed that DENV-2 infection significantly induced COX-2 expression and prostaglandin E-2 (PGE(2)) production in human hepatoma Huh-7 cells. The exogenous expression of COX-2 or PGE2 treatment dose-dependently enhanced DENV-2 replication. In contrast, COX-2 gene silencing and catalytic inhibition sufficiently suppressed DENV-2 replication. In an ICR suckling mouse model, we identified that the COX-2 inhibitor NS398 protected mice from succumbing to life-threatening DENV-2 infection. By using COX-2 promoter-based analysis and specific inhibitors against signaling molecules, we identified that NF-kappa B and MAPK/JNK are critical factors for DENV-2-induced COX-2 expression and viral replication. Altogether, our results reveal that COX-2 is an important factor for DENV replication and can serve as a potential target for developing therapeutic agents against DENV infection.Item type:Item, Deep learning with evolutionary and genomic profiles for identifying cancer subtypes(2019-06-01) ;Lin, Chun-Yu ;Ruan, Peiying ;Li, Ruiming ;Yang, Jinn-MoonSee, SimonCancer subtype identification is an unmet need in precision diagnosis. Recently, evolutionary conservation has been indicated to contain informative signatures for functional significance in cancers. However, the importance of evolutionary conservation in distinguishing cancer subtypes remains largely unclear. Here, we identified the evolutionarily conserved genes (i.e. core genes) and observed that they are primarily involved in cellular pathways relevant to cell growth and metabolisms. By using these core genes, we developed two novel strategies, namely a feature-based strategy (FES) and an image-based strategy (IMS) by integrating their evolutionary and genomic profiles with the deep learning algorithm. In comparison with the FES using the random set and the strategy using the PAM50 classifier, the core gene set-based FES achieved a higher accuracy for identifying breast cancer subtypes. The IMS and FES using the core gene set yielded better performances than the other strategies, in terms of classifying both breast cancer subtypes and multiple cancer types. Moreover, the IMS is reproducible even using different gene expression data (i.e. RNA-seq and microarray). Comprehensive analysis of eight cancer types demonstrates that our evolutionary conservation-based models represent a valid and helpful approach for identifying cancer subtypes and the core gene set offers distinguishable clues of cancer subtypes.Item type:Item, Deep Learning with Evolutionary and Genomic Profiles for Identifying Cancer Subtypes(2018-01-01) ;Lin, Chun-Yu ;Li, Ruiming ;Akutsu, Tatsuya ;Ruan, PeiyingSee, SimonCancer subtype identification is an unmet need in precision diagnosis. The distributed ESD protection scheme was modified to be used in this work. With the consideration of parasitic capacitance from I/O pad, the ESD protection cells have reached the 50-Omega input/output matching to reduce the design complexity for RF circuit designer and to provide suitable ESD protection. Experimental results of these ESD protection cells have successfully verified the ESD robustness and the RF characteristics in the 60-GHz frequency band. These ESD protection cells can easily be used for ESD protection design in the 60-GHz RF applications, and accelerate the design cycle. (C) 2011 Elsevier Ltd. Most of the power is used for converting the signal from digital to analog. In this paper, we proposed a new kind of pixel circuit with fully digital driving. As a result, the signal converting part in the conventional data driver can be omitted, and thus the power consumption can be greatly reduced. The proposed pixel circuit contains two thin-film transistors (TFTs) and a resistor in series, forming a discharging path for pixel voltage. There is also another transistor used to preset the pixel voltage. The gray level is controlled by the pulse width of the data source when the pixel is enabled by the scan voltage. The operation of the novel design is verified by simulation in the cases of a full high-definition panel. The proposed pixel circuit with the new driving method is an innovation approach to reduce data driver power consumption.Item type:Item, Design of 2.4-GHz T/R switch with embedded ESD protection devices in CMOS process(2017-11-01) ;Lin, Chun-Yu ;Liu, Rui-Hong ;Ker, Ming-Dou ;電子工程學系及電子研究所Department of Electronics Engineering and Institute of ElectronicsAs CMOS processes advanced, the integration of radio-frequency (RF) integrated circuits was increasing. In order to protect the fully-integrated RF transceiver from electrostatic discharge (ESD) damage, the transmit/receive (T/R) switch of transceiver frond-end should be carefully designed to bypass the ESD current. This work presented a technique of embedded ESD protection device to enhance the ESD capability of T/R switch. The embedded ESD protection devices of diodes and silicon-controlled rectifier (SCR) are generated between the transistors in T/R switch without using additional ESD protection device. The design procedure of RF circuits without ESD protection device can be simplified. The test circuits of 2.4-GHz transceiver frond-end with T/R switch, PA, and LNA have been integrated and implemented in nanoscale CMOS process to test their performances during RF operations and ESD stresses. The test results confirm that the embedded ESD protection devices can provide sufficient ESD protection capability and it is free from degrading circuit performances. (C) 2017 Elsevier Ltd.
