Item type:Item, A High-Sensitivity CMOS-Compatible Immunosensor Based on the HRP/TMB/Streptavidin System(2008) ;Tai, Yu-Ting ;Chang, Yu-Wei ;Lin, Ming-Yu ;Yang, Yuh-ShyongHuang, Yang-TungIn this report, a CMOS-compatible colorimetric biosensor based on HRP/TMB/streptavidin system is proposed. HRP activity is used for labelled target quantification by photometric detector. The modulation of light intensity due to enzyme reaction can easily be detected in 96-well microarray plate by providing 650nm light source (LED). There is good correlation between absorbance in 650nm and the concentration of HRP-streptavidin with detection limit in 1.33 pM. This device is reusable and suits for various targets by microarray plate modification rather than biochip. With the advantage of portable, low cost, and reusable, this new designed immunosensor could be widely used for both research and diagnostic purposes.Item type:Item, A Miniature CMOS Optoelectronic Biosensor for Screening Skin-Whitening Agents(2009) ;Chang, Yu-Wei ;Lu, Yen-Pei ;Lin, Ming-Yu ;Huang, Yang-TungYang, Yuh-ShyongA miniature CMOS optoelectronic biosensor is proposed for screening whitening agents. Based on the principle of absorption photometry, this biosensor is mainly composed of a green LED, a CMOS phototransistor, and a slide for performing the biochemical reactions. The requirement of the sample volume is only 10 mu l, much less than those in commercial instruments. Melanin assay and MTT assay were performed to evaluate the whitening mechanisms. Experimental results demonstrated that the density of 5x10(4) cells/ml can be successfully detected for both assays. This portable biosensor has great potential toward personalized diagnostic kits for further applications.Item type:Item, Alignment of Stretchable Nanoparticle Chains with Tunable Optical Properties Formed from Molecular Machinery(2013-01-01) ;Lin, Ming-Yu ;Lu, Yen-Pei ;Yang, Yuh-Shyong ;Chen, Hsuen-LiYang, Chih-HuiControl orientation and interparticle spacing of self-assembled nanoparticles can provide versatile functional materials for optical and electronic applications. In this study, we have obtained a glass substratum with tunable optical properties using flowing control and bimolecular competition. The surface of glass was modified from immobilized stretchable gold nanoparticle chains. The long and periodic single-stranded DNA nanotemplates were prepared using Phi29 DNA polymerase molecular machinery according to precise molecular biology regulation. Gold nanoparticle (Au NP) docked at the default sites of the nanotemplates and optical anisotropic absorption were enhanced by approximately 55 % by using tunable, flexible, and oriented one-dimensional metallic nanostructures.Item type:Item, Direct Detection of Long, Periodic, ssDNA nanostructures Assembled on CMOS Transistor Arrays(2009) ;Lin, Ming-Yu ;Chang, Sheng-Ren ;Kao, Jiann-Shiun ;Chen, HsinYang, Yuh-ShyongDNA is a wonderful material for the construction of nanostructures for a variety of applications. This paper describes a novel platform integrating single-stranded DNA (ssDNA) nanotemplates with CMOS-compatible, field-effect sensors. The field-effect sensor was based on oxide-semiconductor field-effect transistors (OSFETs), which were monolithically-integrated with signal-processing circuits to enhance the signal-to-noise ratio, and to detect the construction of ssDNA nanotemplate from complex DNA-protein interactions. The ssDNA nanotemplate on the transistor arrays was prepared by isothermal rolling circle amplification (RCA) through DNA aptamer-protein recognition and self-assembly strategy. The growth of the DNA nanostructure was monitored in situ, real-time and label-free on OSFET.Item type:Item, Facile Synthesis of Radial-Like Macroporous Superparamagnetic Chitosan Spheres with In-Situ Co-Precipitation and Gelation of Ferro-Gels(2012-11-30) ;Yang, Chih-Hui ;Wang, Chih-Yu ;Huang, Keng-Shiang ;Yeh, Chen-ShengWang, Andrew H. -J.Macroporous chitosan spheres encapsulating superparamagnetic iron oxide nanoparticles were synthesized by a facile and effective one-step fabrication process. Ferro-gels containing ferrous cations, ferric cations and chitosan were dropped into a sodium hydroxide solution through a syringe pump. In addition, a sodium hydroxide solution was employed for both gelation (chitosan) and co-precipitation (ferrous cations and ferric cations) of the ferro-gels. The results showed that the in-situ co-precipitation of ferro-ions gave rise to a radial morphology with non-spheroid macro pores (large cavities) inside the chitosan spheres. The particle size of iron oxide can be adjusted from 2.5 nm to 5.4 nm by tuning the concentration of the sodium hydroxide solution. Using Fourier Transform Infrared Spectroscopy and X-ray diffraction spectra, the synthesized nanoparticles were illustrated as Fe3O4 nanoparticles. In addition, the prepared macroporous chitosan spheres presented a super-paramagnetic behaviour at room temperature with a saturation magnetization value as high as ca. 18 emu/g. The cytotoxicity was estimated using cell viability by incubating doses (0 similar to 1000 mu g/mL) of the macroporous chitosan spheres. The result showed good viability (above 80%) with alginate chitosan particles below 1000 mu g/mL, indicating that macroporous chitosan spheres were potentially useful for biomedical applications in the future.Item type:Item, Functionalization, re-functionalization and rejuvenation of ssDNA nanotemplates(2012) ;Lin, Ming-Yu ;Ho, Fu Han ;Yang, Chung-Yao ;Yeh, J. AndrewYang, Yuh-ShyongSingle-stranded DNA (ssDNA) with repetitive sequence was demonstrated to be a versatile nanotemplate for introducing biological activity in a self-assembled manner. Re-functionalization and rejuvenation of the ssDNA nanotemplate were achieved under mild biological conditions without using high temperature and strong alkaline treatment to denature DNA.Item type:Item, Helicity-dependent terahertz emission spectroscopy of topological insulator Sb2Te3 thin films(2017-11-06) ;Tu, Chien-Ming ;Chen, Yi-Cheng ;Huang, Ping ;Chuang, Pei-YuLin, Ming-YuWe report on helicity-dependent terahertz emissions that originate from the helicity-dependent photocurrents in topological insulator Sb2Te3 thin films due to ultrafast optical excitation. The polarity of the emitted terahertz radiation is controlled by both the incident angle and the helicity of optical pulses. Using an unprecedented decomposition-recombination procedure in the time domain, the signals of the Dirac fermions are fully separated from bulk contributions. These results provide insights into the optical coupling of topological surface states and open up opportunities for applying helicity-dependent terahertz emission spectroscopy in spintronics.Item type:Item, Helicity-Dependent Terahertz Radiation from Topological Insulator Sb2Te3 Thin Film by Femtosecond Optical Excitation(2016) ;Tu, Chien-Ming ;Chen, Yi-Cheng ;Huang, Ping ;Chuang, Pei-YuLin, Ming-YuWe report on terahertz radiation from topological insulator (TI)Sb2Te3 thin film under ultrafast optical excitation with different helicity. Polarity-reversals of the emitted THz radiation were observed as the helicity of optical pulses reversed. The observed phenomena are coincident with the characteristics of the helicity-dependent photocurrent on TIs. Our work demonstrates the potential applications of surface photocurrents on TIs for opto-spintronics devices.Item type:Item, Immobilized rolling circle amplification on extended-gate field-effect transistors with integrated readout circuits for early detection of platelet-derived growth factor(2016-07) ;Lin, Ming-Yu ;Hsu, Wen-Yang ;Yang, Yuh-Shyong ;Huang, Jo-WenChung, Yueh-LinDetection of tumor-related proteins with high specificity and sensitivity is important for early diagnosis and prognosis of cancers. While protein sensors based on antibodies are not easy to keep for a long time, aptamers (single-stranded DNA) are found to be a good alternative for recognizing tumor-related protein specifically. This study investigates the feasibility of employing aptamers to recognize the platelet-derived growth factor (PDGF) specifically and subsequently triggering rolling circle amplification (RCA) of DNAs on extended-gate field-effect transistors (EGFETs) to enhance the sensitivity. The EGFETs are fabricated by the standard CMOS technology and integrated with readout circuits monolithically. The monolithic integration not only avoids the wiring complexity for a large sensor array but also enhances the sensor reliability and facilitates massive production for commercialization. With the RCA primers immobilized on the sensory surface, the protein signal is amplified as the elongation of DNA, allowing the EGFET to achieve a sensitivity of 8.8 pM, more than three orders better than that achieved by conventional EGFETs. Moreover, the responses of EGFETs are able to indicate quantitatively the reaction rates of RCA, facilitating the estimation on the protein concentration. Our experimental results demonstrate that immobilized RCA on EGFETs is a useful, label-free method for early diagnosis of diseases related to low-concentrated tumor makers (e.g., PDGF) for serum sample, as well as for monitoring the synthesis of various DNA nanostructures in real time.Item type:Item, Surface composition and interactions of mobile charges with immobilized molecules on polycrystalline silicon nanowires(2015-05-01) ;Lin, Chih-Heng ;Feng, Mei-Huei ;Hwang, Chi-Hung ;Wu, Jennifer Yun-ShinSu, Ping-ChiaThe polycrystalline silicon nanowire field-effect transistor (poly-SiNW FET) is one of the most sensitive sensors used in real-time and label-free biosensing applications. Its low power requirement, mass production potential, and integrability with electronic components make it a highly attractive device in the rapidly growing diagnostic research field. From the viewpoint of device physics, the charges in the vicinity of a nanowire (NW) surface modulate the electrical characteristics of the NW device. The charges can originate from surface molecules or an ionic solution, and their role in NW biosensing systems remains to be clarified. Determining their role is crucial for understanding the physical interactions of charges in a biosensing event and for the realization of NW-based biosensors. Therefore, using poly-SiNW FET, we investigated the interactions of the immobilized molecules on the NW surface with the mobile charges in solutions. We also investigated the interactions between the mobile charged polymers and the immobilized molecules on the NW surface, and we observed the effect of charge neutrality, originating from Coulomb charge-charge interactions, on the NW conductance. Finally, the isoelectric points between native and sulfated PSGL-1 peptides on the NW surface were identified. This study provides a physical understanding of the charge-charge interaction of mobile charges with immobilized charged molecules on a nanoscale surface and presents new opportunities for using charge-based detection in biological and chemical sensing applications. GO was applied for biological applications because of its unique properties, which include water dispensability, exclusive selection of ssDNA, and fluorescence-quenching effects. In this study, aptamer-functionalized GO sheets of nanometer-scale thickness and micrometer-scale dimension were developed. Based on a simple mixed-and-measured strategy, platelet-derived growth factors (PDGF), the tumor marker of ovarian cancer, triggered aptamers conformation switched and deliberated aptamers from GO surface according to thermal dynamic analysis. By functionalizing with various aptamers on GO, the strategy could provide designed functions on GO-based biosensors, bioreactors, and other devices.
