Item type:Item, A Multitheragnostic Nanobubble System to Induce Blood-Brain Barrier Disruption with Magnetically Guided Focused Ultrasound(2015-01-27) ;Huang, Hsin-Yang ;Liu, Hao-Li ;Hsu, Po-Hung ;Chiang, Chih-ShengTsai, Chih-HungItem type:Item, A theranostic nrGO@MSN-ION nanocarrier developed to enhance the combination effect of sonodynamic therapy and ultrasound hyperthermia for treating tumor(2016) ;Chen, Yu-Wei ;Liu, Tse-Ying ;Chang, Po-Hsueh ;Hsu, Po-HungLiu, Hao-LiSonodynamic therapy (SDT), which induces activation of sonosensitizers in cancer cells through ultrasound irradiation, has emerged as an alternative and promising noninvasive therapeutic approach to kill both superficial and deep parts of tumors. In this study, mesoporous silica (MSN) grown on reduced graphene oxide nanosheet (nrGO) capped with Rose Bengal (RB)-PEG-conjugated iron-oxide nanoparticles (IONs), nrGO@MSN-ION-PEG-RB, was strategically designed to have targeted functionality and therapeutic efficacy under magnetic guiding and focused ultrasound (FUS) irradiation, respectively. The singlet oxygen produced by ultrasound-activated RB and the ultrasound-induced heating effect was enhanced by rGO and IONs, which improved the cytotoxic effect in cancer cells. In an animal experiment, we demonstrated that the combination of sonodynamic/hyperthermia therapy with magnetic guidance using this nanocomposite therapeutic agent can produce remarkable efficacious therapy in tumor growth inhibition. Furthermore, the combination effect induced by FUS irradiation produces significant damage to both superficial and deep parts of the targeted tumor.Item type:Item, Comparison of ultrasound temperature imaging with infrared thermometry during radio frequency ablation(2014-04-01) ;Geng, Xiaonan ;Zhou, Zhuhuang ;Li, Qiang ;Wu, ShuicaiWang, Chiao-YinRadio frequency ablation (RFA) is a widely used alternative modality in the treatment of tumors. During RFA, temperature monitoring is essential to ensure accurate and appropriate thermal dosage. Ultrasound temperature imaging based on the detection of echo time-shift has been demonstrated to have good ability to monitor the temperature distribution. However, no study has proven that the region of ultrasound temperature imaging can correspond well to the practical temperature distribution in the tissue. In this study, we aim to combine ultrasound and infrared systems to clarify the correlation between ultrasound temperature imaging and the practical temperature distribution in a tissue. Five porcine livers (n = 5) were ablated using an RFA system and monitored with an ultrasound system to acquire raw backscattered data for temperature imaging. Meanwhile, an infrared imaging system was used to obtain the practical temperature map of the tissue. The results showed that the temperature distribution detected by ultrasound echo time-shift agreed with those obtained from the infrared image. When the tissue temperature was higher than 45 degrees C, ultrasound temperature imaging is difficult to describe the behavior of the heat transfer in a homogeneous medium. In this study, we used the experimental setup based on combining ultrasound and infrared systems to confirm the reliability and limitations of ultrasound temperature imaging in RFA monitoring. Such an experimental design may be considered as an indispensable platform for the development and optimization of ultrasound temperature imaging techniques in RFA monitoring. Nanobubble-based dual contrast enhancement agents encapsulated with perfluoropentane and stabilized with superparamagnetic iron oxide (SPIO) nanoparticles have been synthesized through a single-step emulsion process from thermosensitive F127 and polyacrylic acid (PAA). Both US and MR imaging contrast can be optimized by varying the shell thickness and SPIO-embedded concentration. The US contrast can be enhanced from a mean gray value of 62 to 115, and the MR r(2) value can be enhanced from 164 to 208 (s(-1) mM(-1) Fe) by increasing the SPIO concentration from 14.1 to 28.2 mg/mL, respectively. In vivo investigations of SPIO-embedded nanobubbles in excised tumors under external MT revealed that the US and MR signals change quantitatively compared to the same site without MT. This combined strategy enables the nanobubbles to enhance both passive targeting (increasing the permeability by HIFU) and physical MT of chemotherapeutic drugs to tumors. The integration of functionalities makes this nanobubble system a powerful and viable new tool to achieve simultaneous in vivo tumor imaging and efficacious cancer therapy.
