Item type:Item, Formal Synthesis of 3-Deoxy-D-manno-Octulosonic Acid (KDO) and 3-Deoxy-D-arabino-2-heptulosonic Acid (DAH)(2013-01-01) ;Pradhan, Tapan Kumar ;Lin, Chun Cheng ;Mong, Kwok Kong Tony ;應用化學系Department of Applied ChemistryPractical synthetic routes to 3-deoxy-D-manno-octulo-sonic acid (KDO) and 3-deoxy-D-arabino-2-heptulosonic acid (DAH) from common sugar substrates are reported. Chain homologation of the sugar substrates was accomplished by Wittig olefination and Corey-Fuchs alkynylation. A new cyclization strategy was investigated to access the desired pyranosyl isomer of the KDO target.Item type:Item, Glycosylation Chemistry of 3-Deoxy-D-manno-Oct-2-ulosonic Acid (Kdo) Donors(2015-04-01) ;Pradhan, Tapan Kumar ;Mong, Kwok Kong Tony ;交大名義發表National Chiao Tung University3-Deoxy-D-manno-Oct-2-ulosonic acid (Kdo)-containing oligosaccharides are a conserved carbohydrate component in lipopolysaccharides ( LPSs) of the outer membrane of Gram-negative bacteria, and they are also present in capsular polysaccharides ( CPSs) of bacteria and plant cells. The association of the bacterial LPS with the pathophysiology of bacterial infection have long been recognized. Structure-pathophysiology studies of bacterial infection need to use pure and homogeneous LPS fragments. Such a requirement can be fulfilled by the synthesis of Kdo glycosides and related Kdo-oligosaccharide conjugates. This article provides an overview of the glycosylation chemistries of the synthesis of Kdo alpha-/beta-glycosides, oligosaccharides with Kdo units such as LPS core oligosaccharides, and lipid A derivatives.Item type:Item, Photoluminescent Gold Nanoclusters as Sensing Probes for Uropathogenic Escherichia coli(2013-03-15) ;Chan, Po-Han ;Ghosh, Bhaswati ;Lai, Hong-Zheng ;Peng, Hwei-LingMong, Kwok Kong TonyGlycan-bound nanoprobes have been demonstrated as suitable sensing probes for bacteria containing glycan binding sites. In this study, we demonstrated a facile approach for generating glycan-bound gold nanoclusters (AuNCs). The generated AuNCs were used as sensing probes for corresponding target bacteria. Mannose-capped AuNCs (AuNCs@Mann) were generated and used as the model sensors for target bacteria. A one-step synthesis approach was employed to generate AuNCs@Mann. In this approach, an aqueous solution of tetrachloroauric acid and mannoside that functionized with a thiol group (Mann-SH) was stirred at room temperature for 48 h. The mannoside functions as reducing and capping agent. The size of the generated AuNCs@Mann is 1.95 +/- 0.27 nm, whereas the AuNCs with red photoluminescence have a maximum emission wavelength of similar to 630 nm (lambda(excitation) = 375 nm). The synthesis of the AuNCs@Mann was accelerated by microwave heating, which enabled the synthesis of the AuNCs@Mann to complete within 1 h. The generated AuNCs@Mann are capable of selectively binding to the urinary tract infection isolate Escherichia coli J96 containing the mannose binding protein FimH expressed on the type 1 pili. On the basis of the naked eye observation, the limit of detection of the sensing approach is as low as similar to 2 x 10(6) cells/mL.Item type:Item, Synthesis of Neisseria meningitidis Serogroup W135 Capsular Oligosaccharides for Immunogenicity Comparison and Vaccine Development(2013-08-26) ;Wang, Chia-Hung ;Li, Shiou-Ting ;Lin, Tzu-Lung ;Cheng, Yang-YuSun, Tsung-Hsien
