Item type:Item, Structure and function of TatD exonuclease in DNA repair(2014-09-15) ;Chen, Yi-Chen ;Li, Chia-Lung ;Hsiao, Yu-Yuan ;Duh, YulanderYuan, Hanna S.TatD is an evolutionarily conserved protein with thousands of homologues in all kingdoms of life. It has been suggested that TatD participates in DNA fragmentation during apoptosis in eukaryotic cells. However, the cellular functions and biochemical properties of TatD in bacterial and non-apoptotic eukaryotic cells remain elusive. Here we show that Escherichia coli TatD is a Mg2+-dependent 3\'-5\' exonuclease that prefers to digest single-stranded DNA and RNA. TatD-knockout cells are less resistant to the DNA damaging agent hydrogen peroxide, and TatD can remove damaged deaminated nucleotides from a DNA chain, suggesting that it may play a role in the H2O2-induced DNA repair. The crystal structure of the apo-form TatD and TatD bound to a single-stranded three-nucleotide DNA was determined by X-ray diffraction methods at a resolution of 2.0 and 2.9 angstrom, respectively. TatD has a TIM-barrel fold and the single-stranded DNA is bound at the loop region on the top of the barrel. Mutational studies further identify important conserved metal ion-binding and catalytic residues in the TatD active site for DNA hydrolysis. We thus conclude that TatD is a new class of TIM-barrel 3\'-5\' exonuclease that not only degrades chromosomal DNA during apoptosis but also processes single-stranded DNA during DNA repair.Item type:Item, Expression of an expanded CGG-repeat RNA in a single pair of primary sensory neurons impairs olfactory adaptation in Caenorhabditis elegans(2014-09-15) ;Juang, Bi-Tzen ;Ludwig, Anna L. ;Benedetti, Kelli L. ;Gu, ChenCollins, KimberlyFragile X-associated tremor/ataxia syndrome (FXTAS) is a severe neurodegenerative disorder that affects carriers of premutation CGG-repeat expansion alleles of the fragile X mental retardation 1 (FMR1) gene; current evidence supports a causal role of the expanded CGG repeat within the FMR1 mRNA in the pathogenesis of FXTAS. Though the mRNA has been observed to induce cellular toxicity in FXTAS, the mechanisms are unclear. One common neurophysiological characteristic of FXTAS patients is their inability to properly attenuate their response to an auditory stimulus upon receipt of a small pre-stimulus. Therefore, to gain genetic and cell biological insight into FXTAS, we examined the effect of expanded CGG repeats on the plasticity of the olfactory response of the genetically tractable nematode, Caenorhabditis elegans(C. elegans). While C. elegans is innately attracted to odors, this response can be downregulated if the odor is paired with starvation. We found that expressing expanded CGG repeats in olfactory neurons interfered with this plasticity without affecting either the innate odor-seeking response or the olfactory neuronal morphology. Interrogation of three RNA regulatory pathways indicated that the expanded CGG repeats act via the C. elegans microRNA (miRNA)-specific Argonaute ALG-2 to diminish olfactory plasticity. This observation suggests that the miRNA-Argonaute pathway may play a pathogenic role in subverting neuronal function in FXTAS.Item type:Item, Compositional grading in GaAsSb grown on GaAs substrates(2014-09-15) ;Lin, Y. M. ;Chen, C. H. ;Wu, J. S. ;Lee, C. P.電子工程學系及電子研究所The composition profile and lattice strain of GaAsSb grown on GaAs substrates by molecular beam epitaxy have been investigated using the Rutherford backscattering spectrometry (RBS) and X-ray reciprocal space mapping. Through RBS, we found that the Sb content in the layer increases from the interface to the top surface. The X-ray reciprocal space mapping shows that the GaAsSb lattice also gradually relaxes as the layer becomes thicker. The behavior of composition grading and gradual lattice relaxation is quite different from those of III-V ternary compounds with two group Ill atoms. (C) 2014 Elsevier B.V. H.材料科學與工程學系The La2/3Sr1/3MnO3/BiFeO3 (LSMO/BFO) heterostructure is studied as probed by temperature-dependent ferromagnetic resonance (FMR). Consistent with the structure analysis, two FMR modes are found for both a single LSMO layer and a LSMO/BFO bilayer grown on DyScO3, corresponding to the strained and relaxed phases of LSMO. The FMR linewidth intensity vs temperature curve shows two maxima at 200 K and 140 K in the strained phase of LSMO/BFO bilayer, which is related to the magnetic excitations in BFO. Our results demonstrate a dynamic spin interaction between LSMO and BFO, suggesting an effective route to manipulate the magnetic properties of oxide heterostructures. In the method, an output switching circuit dynamically selects several conversion cells (at least containing know weight conversion cells (KWCC)) as a reference conversion cell group (RCCG) from all conversion cells, and dynamically selects at least one unknown weight conversion cell (UWCC) from all UWCCs. An ADC digitalizes the difference of the output of RCCG and the sum of the outputs of the UWCCs, and inputs the result to a digital controller. The digital controller controls the input of the RCCG according to the output of the ADC to make the output of the RCCG approximate the output of the UWCC. The digital controller uses the outputs of the ADC to work out the actual weights of the UWCCs and stores the actual weights in a calibration memory.Item type:Item, Engineering change order hold time fixing method(2014-09-16) ;Jiang Hiu-Ru ;Yang Yu-MingHo Sung-TingAn ECO hold time fixing method fulfills a short path padding in a placed and routed design by a minimum capacitance insertion. The ex situ RPD AlN nucleation layer can significantly reduce dislocation density and thus improve the crystal quality of the GaN epitaxial layers. Utilizing high-resolution X-ray diffraction, the full width at half maximum of the rocking curve shows that the crystalline quality of the epitaxial layer with the (RPD) AlN nucleation layer is better than that with the low-temperature GaN (LT-GaN) nucleation layer. The threading dislocation density (TDD) is estimated by transmission electron microscopy (TEM), which shows the reduction from 6.8 x 10(7) cm(-2) to 2.6 x 10(7) cm(-2). Furthermore, the light output power (LOP) of the LEDs with the RPD AlN nucleation layer has been improved up to 30 % at a forward current of 350 mA compared to that of the LEDs grown on PSS with conventional LT-GaN nucleation layer.Item type:Item, WIRELESS INTRAOCULAR PRESSURE MONITORING DEVICE, AND DETECTING MODULE THEREOF(2014-09-18) ;Huang Tzuen-Hsi ;Lin Ying-Chun ;Su Wei-Shang ;Cheng Huey-WenHuang Hong-YiA wireless intraocular pressure monitoring device includes reflecting and detecting modules. In this work, we focus on the case where the temporal correlation structure of the fluctuations in the lateral inflow rate can be characterized by the statistics of random fractals. A closed-form expression quantifying the stream flow variability is therefore developed to investigate the influence of the fractal dimension of lateral inflow process and the size of time domain. It is found that the stream flow discharge variability increases with the time domain size, while the fractal dimension of lateral inflow process plays a role in the smoothness of fluctuations in stream flow discharge around the mean.
