Toll-like receptor 9 (TLR9) is the cellular receptor for CpG-ODN in mammalian cells. The CpG-ODN with 18-24 deoxynucleotides that are in current use for human and mouse cells, however, have low activity with rabbit TLR9. Using a cell-based activation assay, we developed a type of CpG-ODN containing a GACGTT or AACGTT motif in 12 phosphorothioate-modified deoxynucleotides with potent stimulatory activity for rabbit TLR9. The developed CpG-ODN have higher activities than other developed CpG-ODN in eliciting antigen-nonspecific immune responses in rabbit splenocytes. When mixed with an NJ85 peptide derived from rabbit hemorrhagic disease virus, they had potent activities to boost an antigen-specific T cell activation and antibody production in rabbits. Compared to Freund\'s adjuvant, the developed CpG-ODN are capable of boosting a potent and less toxic antibody response. The results of this study suggest that both the choice of CpG-motif and its length are important factors for CpG-ODN to effectively activate rabbit TLR9 mediated immune responses.Item type:Item, PEGylation Site-Dependent Structural Heterogeneity Study of MonoPEGylated Human Parathyroid Hormone Fragment hPTH(1-34)(2014-09-30) ;Liu, Chih-Ying ;Li, Xin ;Chen, Wen-Yih ;Chang, Li-ChiaoChen, Yi-FanThe structures of C- and N-terminally monoPEGylated human parathyroid hormone fragment hPTH(1-34) as well as their unmodified counterparts, poly(ethylene glycol) (PEG) and hPTH(1-34), have been studied by small-angle neutron scattering (SANS). The scattering results show that free hPTH(1-34) in 100 mM phosphate buffer (pH 7.4) aggregates into clusters. After conjugation with PEG, the PEG-peptide conjugates self-assemble into a supramolecular core-shell structure with a cylindrical shape. The PEG chains form a shell around the hPTH(1-34) core to shield hPTH(1-34) from the solvent. The detailed structural information on the self-assembled structures is extracted from SANS using a model of the cylindrical core with a shell of Gaussian chains attached to the core surface. On the basis of the data, because of the charge-dipole interactions between the conjugated PEG chain and the peptide, the conjugated PEG chain forms a more collapsed conformation compared to free PEG. Moreover, the size of the self-assembled structures formed by the C-terminally monoPEGylated hPTH(1-34) is about 3 times larger than that of the N-terminally monoPEGylated hPTH(1-34). The different aggregation numbers of the self-assembled structures, triggered by different PEGylation sites, are reported. These size discrepancies because of different PEGylation sites could potentially affect the pharmacokinetics of the hPTH(1-34) drug.Item type:Item, Dynamic reconfigurable heterogeneous processor architecture with load balancing and dynamic allocation method thereof(2014-09-30) ;Chung Chung-Ping ;Yang Hui-ChinChen Yi-ChiA dynamic reconfigurable heterogeneous processor architecture with load balancing and dynamic allocation method thereof is disclosed. The present invention uses a work control logic unit to detect load imbalance between different types of processors, and employs a number of dynamic reconfigurable heterogeneous processors to offload the heavier loaded processors. Hardware utilization of such design can be enhanced, and variation in computation needs among different computation phases can be better handled. To design the dynamic reconfigurable heterogeneous processors, a method of how to choose the basic building blocks and place the routing components is included. With the present invention, performance can be maximized at a minimal hardware cost. Hence the dynamic reconfigurable heterogeneous processor(s) so constructed and the load balancing and dynamic allocation method together will have the best performance at least cost.Item type:Item, High precision patterning of ITO using femtosecond laser annealing process(2014-09-30) ;Cheng, Chung-Wei ;Lin, Cen-Ying ;交大名義發表 ;機械工程學系National Chiao Tung UniversityHigh precision patterning of crystalline indium tin oxide (c-ITO) patterns on amorphous ITO (a-ITO) thin films by femtosecond laser-induced crystallization with a Gaussian beam profile followed by chemical etching is demonstrated. In the proposed approach, the a-ITO thin film is selectively transformed into a c-ITO structure via a low heat affect zone and the well-defined thresholds (ablation and crystallization) supplied by the femtosecond laser pulse. The experimental results show that by careful control of the laser fluence above the crystallization threshold, c-ITO patterns with controllable line widths and ridge-free characteristics can be accomplished. By careful control of the laser fluence above the ablation threshold, fast fabrication of the two parallel sub-micro c-ITO line patterns using a single femtosecond laser beam and single scanning path can be achieved. Along-length sub-micro c-ITO line pattern is fabricated, and the feasibility of fabricating c-ITO patterns is confirmed, which are expected to be used in micro-electronics devices. In this brief, the conventional constant-g(m) biasing is formulated into an analog computation process that calculates the constant-G(m) bias voltage V-0 from an input current. An analog computer measures the effective G(m) from two matched transconductors and converges it to the inverse of a precision resistance by adjusting V-0. Through this, a well-defined large-signal constant-G(m) bias voltage can be found. The proposed technique eliminates the power-law or exponential-law assumptions on the I-out(V-in) characteristics in a conventional design. An interpolation calculation follows to find the optimal small-signal constant-g(m) bias voltage V-out. The fundamental limitations of the proposed technique are the required monotonicity and convexity (or concavity) of the transconductor\'s I-out(V-in). Error sources include mismatches of paired devices and the input offset voltage and open-loop gain of the operation amplifier (OpAmp). These errors are analyzed in detail in this brief. Biasing circuits based on different transistor types are designed using the Taiwan Semiconductor Manufacturing Company (TSMC) 65-nm CMOS process. Computer simulations show that the proposed biasing circuitry can achieve +/- 0.22% g(m) variations from -60 degrees C to 130 degrees C.Item type:Item, Corrugated Surfaces With Tailorable Dark Sectors Within Prescribed Frequency Bands(2014-10-01) ;Kehn, Malcolm Ng Mou ;交大名義發表National Chiao Tung UniversityElectromagnetic bandgap (EBG) surfaces have customarily been defined as being prohibitive of surface-waves within frequency ranges called bandgaps, for all directions along the surface, this latter specification being an understood presumed condition. However, not all practical applications require such an entire bandgap in terms of surface directions. Rather, there could be situations where surface-wave suppression is needed along directions only within an angular sector but are instead desirable towards others outside, over a frequency band, termed herein as a sectoral dark band (SDB). This work investigates this aspect using planar corrugated surfaces as the vehicle for study, in virtue of its ease of surface-wave modal analysis of dispersion by asymptotic corrugations boundary conditions (ACBC). A systematic design procedure for tailoring surfaces to any specified dark sector of weak signals prevailing over any prescribed band is presented.Item type:Item, A highly selective turn-on fluorescence chemosensor for Hg(II) and its application in living cell imaging(2014-10-01) ;Yu, Shiang-Yi ;Wu, Shu-Pao ;應用化學系Department of Applied ChemistryA new coumarin derivative (MS1) containing an imine moiety and a hydroxyl moiety exhibits an enhanced fluorescence in the presence of Hg2+ ions. Other metal ions Al3+, Ca2+, Co2+, Cr3+, Cu2+, Fe2+, Fe3+, Hg-2+,Hg- Mg2+, Mn2+, Ni2+, Pb2+, and Zn2+ produced only minor changes in the fluorescence values of MS1. The binding ratio of MS1-Hg2+ complexes was determined from the Job plot to be 1:1. The binding constant (Ka) of Hg2+ binding to MS1 was found to be 6.85 x 103 M-1. The maximum fluorescence enhancement caused by Hg2+ binding to MS1 was observed in the pH range of 6.5-9.0. Confocal fluorescence microscopy imaging using RAW 264.7 cells showed that MS1 could be used as an effective fluorescent probe for detecting Hg2+ in living cells. (C) 2014 Elsevier B.V. The current research focuses on the determinants of perceived persuasion of the post-purchase arguments and impulsive buyer e-satisfaction. The purpose of this study is to investigate the persuasion of post-purchase arguments on online impulsive buyer satisfaction. A 2 (number of arguments) x 2 (argument strength) experimental design was employed in order to examine whether the persuasion of arguments (e.g. the number and strength of arguments) would vary upon consumers\' tendency to regret. The results indicated that an increased number of arguments provided after the purchase had a stronger positive effect on e-satisfaction when arguments were strong than when arguments were weak. Such an impact would be less pronounced for low tendency-to-regret consumers than for high tendency-to-regret consumers. (C) 2014 Elsevier Ltd. The first type is one of two classical objective functions in scheduling theory: either the total completion time or the makespan. The second type involves an actual cost associated with the processing of a specific job on a given machine: each job-machine combination may have a different cost. Two bi-criteria scheduling problems are considered: (1) minimize the maximum machine cost subject to the total completion time being at its minimum, and (2) minimize the total machine cost subject to the makespan being at its minimum. Since both problems are strongly NP-hard, we propose fast heuristics and establish their worst-case performance bounds. (C) 2014 Elsevier B.V. Here, a scheme using non-uniform sub-bandgap state filling together with intermediate band transport is proposed to resolve the spectral overlap issue. On the basis of detailed balance calculation, spectrally decoupled devices using low-high state filling is shown to achieve 52.8% conversion efficiency when 4eV spectral overlap is present between absorption coefficients of different bands, compared with baseline efficiency equal to 35.1% for conventional half-filled intermediate band devices. If a base material without intermediate band is added to the two section low-high state filling devices, the efficiency is further increased to 61.5%, which approaches efficiency of 63.2% for intermediate band devices with no spectral overlap and 63.8% for unconstrained triple-junction tandem cells. The junction thermalization loss associated with proposed new structures is shown to be equal to conventional half-filled intermediate band devices. S. ;交大名義發表 ;機械工程學系National Chiao Tung UniversityInspired from biology, this study presents a method for designing the fold geometry of deployable reflectors. Since the space available inside rockets for transporting satellites with reflector antennas is typically cylindrical in shape, and its cross-sectional area is considerably smaller than the reflector antenna after deployment, the cross-sectional area of the folded reflector must be smaller than the available rocket interior space. Membrane reflectors in aerospace are a type of lightweight structure that can be packaged compactly. To design membrane reflectors from the perspective of deployment processes, bionic applications from morphological changes of plants are investigated. Creating biologically inspired reflectors, this paper deals with fold geometry of reflectors, which imitate flower buds. This study uses mathematical formulation to describe geometric profiles of flower buds. Based on the formulation, new designs for deployable membrane reflectors derived from bionics are proposed. Adjusting parameters in the formulation of these designs leads to decreases in reflector area before deployment. (C) 2014 IAA. Published by Elsevier Ltd. With only one modulation controller, silicon area and power consumption caused by the replicas of controller can be reduced significantly. Current balance accuracy can be further enhanced since the mismatches between different controllers caused by process, voltage, and temperature variations are removed. Moreover, the offset cancellation control embedded in the current matching unit is used to eliminate intrinsic offset voltage existing at the operational transconductance amplifier for improved current balance. An explicit model, which contains both voltage and current balance loops with non-ideal effects, is derived for analyzing system stability. Experimental results show that current difference between each phase can be decreased by over 83% under both heavy and light load conditions.Item type:Item, Modeling team performance and its determinants in high-tech industries: Future trends of virtual teaming(2014-10-01) ;Joe, Sheng-Wuu ;Tsai, Yuan-Hui ;Lin, Chieh-Peng ;Liu, Wei-Te經營管理研究所Drawing upon social cognitive theory, this research postulates critical team characteristics and environmental factors as key drivers of team performance in virtual contexts. In the proposed model of this study, team performance is positively related to four antecedents (i.e., team efficacy, collective outcome expectation, emotional intelligence climate, and procedural justice climate) directly and indirectly via the mediation of team planning. An empirical testing of this research, by investigating working professionals in high-tech industries, suggests the applicability of social cognitive theory in understanding team performance in virtual teaming contexts. Last, managerial implications about future virtual teaming and limitations of this research are provided. (C) 2014 Elsevier Inc. The GZO nanorods with well aligned and extremely dense properties are considered as a thin film for RRAM device. The oxygen vacancies can be confined and migrate along grain boundaries in the GZO nanorod film. Therefore, the weakest point for formation and rupture of conductive filament can be limited at the interface between GZO nanorod film and ZnO seeding layer. Compared with ITO/ZnO/ITO device, a significant improvement in the distribution of high resistance state (HRS) and low resistance state (LRS) during resistance switching is demonstrated in the present device. In addition, a high endurance of more than 7000 cycles with the resistance ratios of HRS/LRS about 200 times is achieved in this device. The ITO/GZO/ZnO/ITO device is a good candidate for the transparent RRAM application.
