Item type:Item, The First Residue of the PWWP Motif Modulates HATH Domain Binding, Stability, and Protein-Protein Interaction(2015-07-07) ;Hung, Yi-Lin ;Lee, Hsia-Ju ;Jiang, Ingjye ;Lin, Shang-ChiLo, Wei-ChengHepatoma-derived growth factor (hHDGF) and HDGF-related proteins (HRPs) contain conserved N-terminal HATH domains with a characteristic structural motif; namely the PWWP motif. The HATH domain has attracted attention because of its ability to bind with heparin/heparan sulfate, DNA, and methylated histone peptide. Depending on the sequence of the PWWP motif, HRP HATHs are classified into P-type (Pro-His-Trp-Pro) and A-type (Ala-His-Trp-Pro) forms. A-type HATH is highly unstable and tends to precipitate in solution. We replaced the Pro residue in P-type HATH(HDGF) with Ala and evaluated the influence on structure, dynamics, and ligand binding. Nuclear magnetic resonance (NMR) hydrogen/deuterium exchange and circular dichroism (CD) measurements revealed reduced stability. Analysis of NMR backbone N-15 relaxations (R-1, R-2, and nuclear Overhauser effect) revealed additional backbone dynamics in the interface between the beta-barrel and the C-terminal helix bundle. The beta 1-beta 2 loop, where the AHWP sequence is located, has great structural flexibility, which aids HATH-HATH interaction through the loop. A-type HATH, therefore, shows a stronger tendency to aggregate when binding with heparin and DNA oligomers. This study defines the role of the first residue of the PWWP motif in modulating HATH domain stability and oligomer formation in binding.Item type:Item, Importance of Internal Porosity for Glucan Adsorption in Mesoporous Carbon Materials(2015-07-07) ;Chung, Po-Wen ;Charmot, Alexandre ;Click, Timothy ;Lin, YuchunBae, YounJueTo better understand the adsorption of long-chain poly(1 -> 4)beta-D-glucans on carbon surfaces as well as interactions responsible for this adsorption, we use a comparative study involving mesoporous carbon silica composite materials that have been etched to varying degrees and all-atom molecular dynamics simulations. The materials synthesized as part of this etching study consist of an as-synthesized composite material (MCN-MSN), MCN-MSN-0.5 (composite materials consisting of 50% carbon by mass), MCN-MSN-0.3 (composite materials consisting of 70% carbon by mass), and MCN, in which silica etching was conducted using an aqueous ethanolic solution of either NaOH or HF. Data for the adsorption of long-chain glucans to these materials from concentrated aqueous HCl (37 wt %) solution demonstrate a direct relationship between the amount of beta-glu adsorption and the magnitude of exposed carbon mesopore surface area, which systematically increases and is also accompanied by an increase in the mesopore size during silica etching. This demonstrates beta-glu adsorption as occurring on internal carbon mesopores rather than exclusively on the external carbon surface. These experimental data on adsorption were corroborated by molecular dynamics (MD) simulations of beta-glu adsorption to a graphene bilayer separated by a distance of 3.2 nm, chosen to correspond to the carbon mesopore diameter of the experimental system. Simulation results using a variety of beta-glu solvent systems demonstrate the rapid adsorption of a beta-glu strand on the graphitic carbon surface via axial coupling and are consistent with experimentally observed trends in fast adsorption kinetics. Solvent-mediated effects such as small-scale hydrophobicity and preferential interactions with ions are shown to play important roles in modulating glucan adsorption to carbon surfaces, whereas experimental data on hydrophobically modified silica demonstrate that hydrophobicity in and of itself is insufficient to cause beta-glu adsorption from concentrated aqueous HCl solution.Item type:Item, Ovarian Suspension With Adjustable Sutures: An Easy and Helpful Technique for Facilitating Laparoendoscopic Single-Site Gynecologic Surgery(2015-07-08) ;Chen, Kuo-Hu ;Chen, Li-Ru ;Seow, Kok-Min ;機械工程學系Department of Mechanical EngineeringStudy Objective: To describe a method of ovarian suspension with adjustable sutures (OSAS) for facilitating laparoendoscopic single-site gynecologic surgery (LESS) and to investigate the effect of OSAS on LESS. Design: Prospective cohort study (Canadian Task Force classification: II-2). Setting: University teaching hospital. Patients: One hundred seventy-eight patients with benign 5-to 15-cm cystic ovarian tumors who underwent LESS with OSAS (suspension group, n = 90) and without OSAS (control group, n = 88). Interventions: For patients who underwent OSAS (suspension group), 1 end of double-head straight needles with a polypropylene suture was inserted into the pelvic cavity through the abdominal skin to penetrate the cyst or ovarian parenchyma and puncture outside the abdominal skin. After cutting off the needles, both sides of the remaining suture were held together by a clamp, without knotting, so that the manipulator could "lift," "loosen," or "fix" the stitches to adjust the tension. Measurements and Main Results: The average time to create OSAS was 2.9 min. For the suspension and control groups, the average blood loss was 81.4 and 131.8 mL (p < .001), and the operative time was 42.0 and 61.3 min (p < .001), respectively. There were no significant differences in the incidence of complications (5.6% vs 9.1%; p = .365), but there were significant differences in conversions to standard non single-site laparoscopy (5.6% vs 15.9%; p = .025) and laparotomy (1.1% vs 6.8%; p = .040). Logistic regression analysis revealed that the ratios of conversion to standard non single-site laparoscopy (odds ratio [On 0.126; 95% confidence interval [C1], 0.311-0.508) and laparotomy (OR, 0.032; 95% CI, 0.002-0.479) were much lower in the suspension group; the risk of complications was comparable (OR, 0.346; 95% CI, 0.085-1.403). Conclusion: OSAS is an easy, safe, and feasible method that offers advantages during LESS. Although routine use of OSAS is not necessary, OSAS can be considered during LESS to facilitate the surgery. Duran ;Ng, Tien KheeTsai, Cheng-TingWith increasing interest in visible light communication, the laser diode (LD) provides an attractive alternative, with higher efficiency, shorter linewidth and larger bandwidth for high-speed visible light communication (VLC). Previously, more than 3 Gbps data rate was demonstrated using LED. By using LDs and spectral-efficient orthogonal frequency division multiplexing encoding scheme, significantly higher data rates has been achieved in this work. Using 16-QAM modulation scheme, in conjunction with red, blue and green LDs, data rates of 4.4 Gbps, 4 Gbps and 4 Gbps, with the corresponding BER/SNR/EVM of 3.3 x 10(-3)/15.3/17.9, 1.4 x 10-3/16.3/15.4 and 2.8 x 10(-3)/15.5/16.7 were obtained over transmission distance of similar to 20 cm. We also simultaneously demonstrated white light emission using red, blue and green LDs, after passing through a commercially available diffuser element. Our work highlighted that a tradeoff exists in operating the blue LDs at optimum bias condition while maintaining good color temperature. The best results were obtained when encoding red LDs which gave both the strongest received signal amplitude and white light with CCT value of 5835K. Aliasing due to the lenslet arrays is one of the limitations of InIm displays. In this paper, we propose a dynamic InIm display using electrically movable liquid crystal (LC) lens array to implement the moving array lenslet technique (MALT) and to eliminate the multifacet phenomenon in the 3-D images. The improvement of the viewing quality of dynamic InIm display is experimentally verified. A low-power noise-like pulse train generated by a ring oscillator was amplified using a two-stage amplifier, where the performance of the second-stage amplifier determined the final output power level. The optical intensity in the second-stage amplifier was managed well to avoid not only the excessive spectral broadening induced by nonlinearities but also any damage to the device. On the other hand, the power conversion efficiency of the amplifier was optimized through proper control of its pump wavelength. The pump wavelength determines the pump absorption and therefore the power conversion efficiency of the gain fiber. Through this approach, the average power of the noise-like pulse train was amplified considerably to an output of 13.1 W, resulting in a power conversion efficiency of 36.1% and a pulse energy of 0.85 mu J. To the best of our knowledge, these amplified pulses have the highest average power and pulse energy for noise-like pulses in the 1.56-mu m wavelength region. As a result, the net gain in the cascaded amplifier reached 30 dB. With peak and pedestal widths of 168 fs and 61.3 ps, respectively, for the amplified pulses, the pedestal-to-peak intensity ratio of the autocorrelation trace remains at the value of 0.5 required for truly noise-like pulses. By X-ray diffraction measurements, different types of SmCG (B8) phases and leaning angles were controlled by the shape effect of the string- and cage-like siloxane termini for Si and P1 (with only one arm of H-bonded bent-core), respectively. In addition, the confinement effect of Pi, C4, and P8 (accompanied by increasing the numbers of attached H-bonded bent-core arms) resulted in higher transition temperatures and the diminishing of mesophasic ranges (even the disappearance of mesophase in P8). Moreover, AFM images showed the bilayer smectic CG phases of Si and P1 were aligned to reveal highly ordered thread-like structures by a DC field. By spontaneous polarization measurements within the mesophasic ranges, Si and P1 showed ferroelectric transitions but C4 displayed antiferroelectricity. Finally, the electro-optical performance of B8 phases could be optimized through binary mixtures of Si and P1, and a well aligned modulated ribbon phase could be formed via specific molar ratios of the binary mixtures.
