This BG-SAM possesses saturation fluence of 108.3 mu J/cm(2), modulation depth (Delta R) of 6.5%, non-saturable loss of 38.4%, high damage threshold above 1.354 mJ/cm(2) and excellent uniformity providing for the generation of passive mode-locked (ML) pulses for erbium-doped fiber lasers (EDFLs) on a large sample area. Under 124 mW 976 nm pumping, We obtained 452-fs continuous-wave ML pulses with pulse energy of 91 pJ and full width at halfmaximum (FWHM) of 6.72-nm from this EDFL. The results clearly evidence that the PLD is an efficient method for fabricating BG-SAM that is suitable for a compact ultrafast ML fiber laser system.Item type:Item, In-Oleo Microgasometry of Nanoliter-Scale Gas Volumes with Image-Based Detection(2016-12-06) ;Prabhu, Gurpur Rakesh D. ;Urban, Pawel L. ;應用化學系Department of Applied ChemistryGasometric assays involve measurements of the amounts of gases that are released during physical or chemical processes. The available instrumentation for gasometric analysis is generally difficult to use and requires large sample volumes. In some cases, toxic materials (mercury) are involved in the analysis process. Here, we propose a microscale gasometric assay using silicone oil as matrix. Microliter-volume (similar to 2.5 mu L) aqueous droplets, containing sample and reagent and/or catalyst, are introduced to the oil matrix and merged. Nanoliters of gaseous products are released to the surrounding oil matrix forming tiny spherical bubbles. Due to the huge differences between refractive indices of the released gas and the surrounding liquids (aqueous assay solution, oil), the gas bubbles are clearly visible from the top, when the assay reservoir is illuminated from the bottom with light emitting diodes. The released gas bubbles are documented by recording videos of the assay reservoir. Individual frames within these videos are then analyzed by a graphical software to obtain diameters of every gas bubble at each time point. Following a fixed period of time (typically, 5-90 s) after the sample/reagent droplet merger, the volume of the released gas scales with the amount of the substrate (analyte) present in the sample droplet. For example, hydrogen peroxide can be decomposed to oxygen by 0.44 U catalase enzyme and semiquantified in the range up to similar to 1.0 mu mol. Glutathione can be detected in a two-step procedure ((1) oxidation of glutathione by hydrogen peroxide; (2) decomposition of the hydrogen peroxide residue by catalase).Item type:Item, Plug-Volume-Modulated Dilution Generator for Flask-Free Chemistry(2016-12-06) ;Liu, Pei-Han ;Urban, Pawel L. ;應用化學系 ;應用化學系分子科學碩博班Department of Applied ChemistryDilution is one of the common chemical procedures which are carried out in all chemistry laboratories to prepare standard solutions with different concentrations for assay calibration, and to reduce matrix effects while handling complex samples. Yet dilution is mostly performed manually using large volume manual liquid-handling tools (volumetric flasks, graduated cylinders, and pipettes). Here we describe a simple continuous and automated method of diluting complex samples and stock solutions using an Arduino-based control unit. The proposed plug-volume-modulation approach relies on continuous introduction of short plugs of samples separated with short plugs of solvent. The train of sample pulses is generated by opening and closing two pinch valves interchangeably, so that either sample or solvent can enter the mixing zone (T-junction). The plugs of sample and solvent are pulled along a transfer flow line by a peristaltic pump. They mix due to advection, turbulence, and diffusion. The effluent of the flow line supplies diluted samples with well-defined dilution factors. The desired dilution factor is programmed by setting the duration of sample and solvent pulses injected in every cycle. The sample duty cycle effectively determines the dilution factor. Initially, we verified the quality of the generated diluted samples by off-line and online optical detection. We further demonstrated the usefulness of this dilution generator when selecting the optimum dilution factors for complex samples analyzed by direct infusion electrospray ionization mass spectrometry. The proposed method eliminates conventional glassware from dilution steps. Hence, it can readily be incorporated into automated analytical systems.Item type:Item, Further resolution enhancement of high-sensitivity laser scanning photothermal microscopy applied to mouse endogenous(2016-12-07) ;Nakata, Kazuaki ;Tsurui, Hiromichi ;Kobayashi, Takayoshi ;電子物理學系Department of ElectrophysicsPhotothermal microscopy has intrinsically super resolution capability due to the bilinear dependence of signal intensity of pump and probe. In the present paper, we have made further resolution improvement of high-sensitivity laser scanning photothermal microscopy by applying non-linear detection. By this, the new method has the following advantages: (1) super resolution with 61% and 42% enhancement from the diffraction limit values of the probe and pump wavelengths, respectively, by a second-order non-linear scheme, (2) compact light source using inexpensive conventional diode lasers, (3) wide applicability to nonfluorescent materials such as gold nanoparticles (GNPs) and hematoxylin-eosin stained biological samples, (4) relative robustness to optical damage, and (5) a high-frame rate using a Galvano mirror. The maximum resolution is determined to be 160 nm in the second-order non-linear detection mode and 270 nm in the linear detection mode by the PT signal of GNPs. The pixel rate and frame rate for 300 x 300 pixel image are 50 mu s and 4.5 s, respectively. The pixel and frame rate are shorter than the rates, which are 1ms and 100 s, respectively, using the piezo-driven stage system. The highest occupied molecular orbital (HOMO) and lowest unoccupied (LUMO) molecular orbital energy levels of these terpolymers indicate that there is sufficient LUMO offset with PCBM for efficient exciton dissociation, and their deeper HOMO levels ensure the high open-circuit voltage for the resultant bulk heterojunction solar cells. Measurements on the solar cell devices also confirm that compared to those based on P2 the devices based on P1 possess a higher short-circuit photocurrent (J(sc)) as well as a higher fill factor (FF), which is attributed to the lower bandgap and higher hole mobility for P1, whereas the V-oc is higher for the devices that are based on P2, which may be a result of P2 having a lower HOMO energy level than P1. The optimized polymer solar cells fabricated using P1:PC71BM (DIO/CF) and P2:PC71BM (CF/DIO) for the active layers showed a PCE of 7.19% and 6.34%, respectively. Atomic force microscopy (AFM) images of PEPC71BM blend films show that they exhibit more suitable morphology with favorable interpenetrating networks, which favors high J(sc) and FF. Moreover, P1 exhibits a more crystalline nature than P2 that also favors the charge transport. This may be a result of better molecular packing, more distinct phase separation of the blended films, as well as a reduction of charge recombination.
