Item type:Item, Inhibition effect of curcumin on UVB-induced secretion of pro-inflammatory cytokines from corneal limbus epithelial cells(2017-06-18) ;Chao, Shih-Chun ;Hu, Dan-Ning ;Roberts, Joan ;Shen, XilunLee, Chia-YiAIM: To study the effects of curcumin on the secretion of interleukin (IL) -6 and IL-8 by corneal limbus epithelial cells. METHODS: Human corneal limbus epithelial cells were isolated and cultured from donor eyes and irradiated by UVB at different dosages with or without curcumin. MTT test was used for studying the effects of UVB and curcumin on the cell viability. The role of mitogen-activated protein kinase (MAPK) and nuclear factor-kappa B (NF-kappa B) pathways on the UVB-induced secretion of IL-6 and IL-8 were tested by addition of their inhibitors to the culture with or without UVB-radiation. Levels of various signal pathways, IL-6 and IL-8 in the cells and in the conditioned culture medium were measured by ELISA analysis. RESULTS: UVB at 20 mJ/cm(2) or less and curcumin at 20 mu mol/L or less did not affect the cell viability of cultured limbus epithelial cells (P>0.05). UVB irradiation at 10 and 20 mJ/cm(2) induced a significant increase of secretion of IL-6 and IL-8 and upregulated NF-kappa B and phosphorylated MAPK pathways of cultured limbus epithelial cells (P<0.05). Various signal pathway inhibitors, including SP600125 (JNK inhibitor), SB203580 (p38 MAPK inhibitor) and BAY11-7082 (NF-kappa B inhibitor) significantly decreased the UVB-induced secretion of IL-6 and IL-8 secretion (P<0.05). Curcumin at 5-20 mu mol/L significantly inhibited UVB-induced secretion of IL-6 and IL-8 by limbus epithelial cells in a dose-dependent manner; while curcumin alone did not affect the secretion of IL-6 and IL-8. The upregulation of NF-kappa B and MAPK pathways induced by UVB treatment was significantly inhibited by curcumin, suggesting that NF-kappa B and MAPK pathways are involved in the inhibitory effect of curcumin on UVB-induced production of IL-6 and IL-8. CONCLUSION: Curcumin may be a promising agent to be explored for the prevention and treatment of pterygium.Item type:Item, Evolution of the Reactor Antineutrino Flux and Spectrum at Daya Bay(2017-06-19) ;An, F. P. ;Balantekin, A. B. ;Band, H. R. ;Bishai, M.Blyth, S.The Daya Bay experiment has observed correlations between reactor core fuel evolution and changes in the reactor antineutrino flux and energy spectrum. Four antineutrino detectors in two experimental halls were used to identify 2.2 million inverse beta decays (IBDs) over 1230 days spanning multiple fuel cycles for each of six 2.9 GW(th) reactor cores at the Daya Bay and Ling Ao nuclear power plants. Using detector data spanning effective Pu-239 fission fractions F-239 from 0.25 to 0.35, Daya Bay measures an average IBD yield (sigma) over bar (f) of (5.90 +/- 0.13) x 10(-43) cm(2)/fission and a fuel-dependent variation in the IBD yield, d sigma(f)/dF(239), of (-1.86 +/- 0.18) x 10(-43) cm(2)/fission. This observation rejects the hypothesis of a constant antineutrino flux as a function of the Pu-239 fission fraction at 10 standard deviations. The variation in IBD yield is found to be energy dependent, rejecting the hypothesis of a constant antineutrino energy spectrum at 5.1 standard deviations. While measurements of the evolution in the IBD spectrum show general agreement with predictions from recent reactor models, the measured evolution in total IBD yield disagrees with recent predictions at 3.1 sigma. This discrepancy indicates that an overall deficit in the measured flux with respect to predictions does not result from equal fractional deficits from the primary fission isotopes U-235, Pu-239, U-238, and Pu-241. Based on measured IBD yield variations, yields of (6.17 +/- 0.17) and (4.27 +/- 0.26) x 10(-43) cm(2)/fission have been determined for the two dominant fission parent isotopes U-235 and Pu-239. A 7.8% discrepancy between the observed and predicted U-235 yields suggests that this isotope may be the primary contributor to the reactor antineutrino anomaly.Item type:Item, Schiff Base Proton Acceptor Assists Photoisomerization of Retinal Chromophores in Bacteriorhodopsin(2017-06-20) ;Hung, Chih-Chang ;Chen, Xiao-Ru ;Ko, Ying-Kuan ;Kobayashi, TakayoshiYang, Chii-ShenIn this study, we investigated the ultrafast dynamics of bacteriorhodopsins (BRs) from Haloquadratum walsbyi (HwBR) and Haloarcula marismortui (HmBRI and HmBRII). First, the ultrafast dynamics were studied for three HwBR samples: wild-type, D93N mutation, and D104N mutation. The residues of the D93 and D104 mutants correspond to the control by the Schiff base proton acceptor and donor of the proton translocation subchannels. Measurements indicated that the negative charge from the Schiff base proton acceptor residue D93 interacts with the ultrafast and substantial change of the electrostatic potential associated with chromophore isomerization. By contrast, the Schiff base proton donor assists the restructuring of the chromophore cavity hydrogen-bond network during the thermalization of the vibrational hot state. Second, the ultrafast dynamics of the wild-types of HwBR, HmBRI, and HmBRII were compared. Measurements demonstrated that the hydrogen-bond network in the extracellular region in HwBR and HmBRII slows the photoisomerization of retinal chromophores, and the negatively charged helices on the cytoplasmic side of HwBR and HmBRII accelerate the thermalization of the vibrational hot state of retinal chromophores. The similarity of the correlation spectra of the wild-type HmBRI and D104N mutant of HwBR indicates that inactivation of the Schiff base proton donor induces a positive charge on the helices of the cytoplasmic side.Item type:Item, 交大e-News第234期-美國輝達公司(NVIDIA)創辦人暨執行長黃仁勳 獲頒交大名譽博士(2017-06-20) ;國立交通大學秘書室SecretariatItem type:Item, High efficiency ternary organic solar cell with morphology-compatible polymers(2017-06-21) ;Mai, Jiangquan ;Lu, Haipeng ;Lau, Tsz-Ki ;Peng, Shih-HaoHsu, Chain-ShuTernary organic solar cell is a promising strategy for improving the power conversion efficiency (PCE) of single-junction organic solar cells by broadening the light absorption spectrum with the incorporation of two electron donors and one acceptor or one donor and two acceptors. However, in many cases the optimized loading of the third component is so small that its contribution to the light absorption is limited. Following our previously proposed selection rule for the two polymers capable of comparable loadings, we report in this work a ternary system composed of two morphologically compatible polymers with distinct chemical structures that can achieve a high PCE of 9.0% with a mass ratio of 1:1. Besides the expected improvement in short circuit current due to the broadening of the absorption spectrum, the fill factor of the ternary device is improved significantly. We attribute it to the balanced electron and hole mobility and reduced recombination benefiting from the high morphology compatibility of the system.Item type:Item, Blending Homopolymers for Controlling the Morphology Transitions of Block Copolymer Nanorods Confined in Cylindrical Nanopores(2017-06-21) ;Cheng, Ming-Hsiang ;Hsu, Yu-Chen ;Chang, Chun-Wei ;Ko, Hao-WenChung, Pei-YunThe microphase separation of block copolymers in confined geometries has been widely investigated over the last few decades. The controllability and versatility of the confinement-induced morphologies, however, are still difficult to be achieved because of the limited experimental parameters in the process of fabricating the confined nanostructures. In this work, we study the morphology transitions of lamellae forming polystyrene-block-polydimethylsiloxane (PS-b-PDMS) nanorods confined in the nanopores of anodic aluminum oxide (AAO) templates. The nanorods are formed by solvent-assisted template wetting, and the morphologies are compared to those in the bulk state. By blending PS-b-PDMS with homopolystyrene (hPS), the morphologies of the nanorods can be controlled because of the changes of the effective volume fractions. Special morphology transitions from concentric lamellar morphology, to multihelical morphology, and finally to spherical-like morphology are observed by increasing the weight ratios of hPS. hPS with different molecular weights is also applied to investigate the effect of hPS on the morphologies of the PS-b-PDMS/hPS blend nanostructures. The unusual morphologies are further confirmed by a selective removal process, which also generates nanochannels for possible refilling with functional materials.Item type:Item, Understanding the functional properties of bio-inorganic nanoflowers as biocatalysts by deciphering the metal-binding sites of enzymes(2017-06-21) ;Escobar, Sindy ;Velasco-Lozano, Susana ;Lu, Chih-Hao ;Lin, Yu-FengMesa, MonicaThe biomineralisation of metal phosphates is a promising approach to develop more efficient nano-biocatalysts; however, the interactions between the protein and the inorganic mineral are poorly understood. Elucidating which protein regions most likely participate in the mineral formation will guide the fabrication of more efficient biocatalysts based on metal-phosphate nanoflowers. We have biomineralised the lipase from Thermomyces lanuginosus using three calcium, zinc and copper phosphates to fabricate different types of bio-inorganic nanoflowers. To better understand how the biomineralisation process affects the enzyme properties, we have computationally predicted the protein regions with a higher propensity for binding Ca2+, Cu2+ and Zn2+. These binding sites can be considered as presumable nucleation points where the biomineralisation process starts and explain why different metals can form bio-inorganic nanoflowers of the same enzyme with different functional properties. The formation of calcium, copper and zinc phosphates in the presence of this lipase gives rise to nanoflowers with different morphologies and different enzymatic properties such as activity, stability, hyperactivation and activity-pH profile; these functional differences are supported by structural studies based on fluorescence spectroscopy and can be explained by the different locations of the predicted nucleation sites for the different metals. Among the three metals used herein, the mineralisation of this lipase with zinc-phosphate enables the fabrication of bio-inorganic nanoflowers 34 times more stable than the soluble enzyme. These bio-inorganic nanoflowers were reused for 8 reaction cycles achieving 100% yield in the hydrolysis of p-nitrophenol butyrate but losing more than 50% of their initial activity after 6 operational cycles. Finally, this heterogeneous biocatalyst was more active and enantioselective than the soluble enzyme (ee = 79%(R)) towards the kinetic resolution of rac-1-phenylethyl acetate yielding the R enantiomer with ee = 84%.Item type:Item, METHOD OF PREPARING SELECTIVE CATALYTIC REDUCTION COMPOSITE CATALYST(2017-06-22) ;Hsun-Ling Bai ;Liang-Yi Lin ;Tsung-Yu LeeYou-Ren ZhangA method of preparing selective catalytic reduction composite catalyst is provided. Absorption enhancement was observed extending 1-2 orders of magnitude over that of a reference TiO2 film. Several enhancement peaks in the region from 600-950 nm were identified, which far exceed both the ergodic fundamental limit and the limit based on surface-gratings, with some peaks exceeding 100 times enhancement. These results are attributed to radically sharp refraction where the optical path length approaches infinity due to the Poynting vector lying nearly parallel to the photonic crystal interface. The observed phenomena follow directly from the simple cubic symmetry of the photonic crystal, and can be achieved by integrating the light-trapping architecture into the absorbing volume. These results are not dependent on the material used, and can be applied to any future light trapping applications such as phosphor-converted white light generation, water-splitting, or thin-film solar cells, where increased response in areas of weak absorption is desired.Item type:Item, A rapid and quantitative method to detect human circulating tumor cells in a preclinical animal model(2017-06-23) ;Tu, Shih-Hsin ;Hsieh, Yi-Chen ;Huang, Li-Chi ;Lin, Chun-YuHsu, Kai-WenBackground: As cancer metastasis is the deadliest aspect of cancer, causing 90% of human deaths, evaluating the molecular mechanisms underlying this process is the major interest to those in the drug development field. Both therapeutic target identification and proof-of-concept experimentation in anti-cancer drug development require appropriate animal models, such as xenograft tumor transplantation in transgenic and knockout mice. In the progression of cancer metastasis, circulating tumor cells (CTCs) are the most critical factor in determining the prognosis of cancer patients. Several studies have demonstrated that measuring CTC-specific markers in a clinical setting (e.g., flow cytometry) can provide a current status of cancer development in patients. However, this useful technique has rarely been applied in the real-time monitoring of CTCs in preclinical animal models. Methods: In this study, we designed a rapid and reliable detection method by combining a bioluminescent in vivo imaging system (IVIS) and quantitative polymerase chain reaction (QPCR)-based analysis to measure CTCs in animal blood. Using the IVIS Spectrum CT System with 3D-imaging on orthotropic-developed breast-tumor-bearing mice. Results: In this manuscript, we established a quick and reliable method for measuring CTCs in a preclinical animal mode. The key to this technique is the use of specific human and mouse GUS primers on DNA/RNA of mouse peripheral blood under an absolute qPCR system. First, the high sensitivity of cancer cell detection on IVIS was presented by measuring the luciferase carried MDA-MB-231 cells from 5 to 5x10(11) cell numbers with great correlation (R-2 = 0.999). Next, the MDA-MB-231 cell numbers injected by tail vein and their IVIS radiance signals were strongly corrected with qPCR-calculated copy numbers (R-2 > 0.99). Furthermore, by applying an orthotropic implantation animal model, we successfully distinguished xenograft tumor-bearing mice and control mice with a significant difference (p < 0.001), whereas IVIS Spectrum-CT 3D-visualization showed that blood of mice with lung metastasis contained more than twice the CTC numbers than ordinary tumor-bearing mice. We demonstrated a positive correlation between lung metastasis status and CTC numbers in peripheral mouse blood. Conclusion: Collectively, the techniques developed for this study resulted in the integration of CTC assessments into preclinical models both in vivo and ex vivo, which will facilitate translational targeted therapy in clinical practice.Item type:Item, Effects of substrate and annealing on GaN films grown by plasma-assisted molecular beam epitaxy(2017-06-25) ;Yang, Zu-Po ;Tsou, Tsung-Han ;Lee, Chao-Yu ;Kan, Ken-YuanYu, Ing-SongGaN films were deposited by plasma-assisted molecular beam epitaxy on different substrates, i.e. Si (111), Sapphire (0001) and 4H-SiC (0001) with 4 degrees of miscutting orientation. Due to the lattice mismatch between GaN films and substrates, the substrates profoundly affect the growth mechanism of GaN films on them during the epitaxy. The smooth and complete GaN films can be obtained on 4H-SiC substrates based on the analyses of reflection high energy electron diffraction, scanning electron microscopy and atomic force microscopy. Higher quality of GaN films on 4H-SiC was also confirmed by high resolution X-ray diffraction. Photoluminescence measurements show that the compress stress existed in GaN films grown on sapphire, but tensile stress was observed in the films grown on 4H-SiC and Si. Meanwhile, in-situ annealing treatment of post expitxy can remove Ga droplets on GaN films and decrease the surface roughness and defect density of GaN films; however, thermal decomposition on the surface of GaN films was observed in the characterizations of X-ray photoelectron spectroscopy. The composite TCLs were fabricated from aluminum-doped zinc oxide (AZO) layers grown through atomic-layer deposition and pulsed-laser deposition. The results showed that wet-etching of composite AZO thin films improved the light extraction efficiency of the corresponding LED devices. The droop efficiencies of LEDs with flat AZO thin films, composite AZO thin films, and optimum textured composite AZO thin films were 29%, 30%, and 22%, respectively. LEDs with textured composite AZO layers exhibited favorable optoelectronic performance. When the injection current was increased from 20 to 1000 mA, the emission wavelengths of LEDs with flat AZO thin films, composite AZO thin films, and optimum textured composite AZO thin films shifted by 3, 5, and 3 nm, respectively. The optimum textured composite AZO thin film can not only improve LED performance but also effectively substitute flat TCLs in AlGaInP LED applications. (C) 2016 Elsevier B.V. Two types of via-contact structure TFTs are investigated: symmetrical and UI structure (TFTs with Iand U-shaped asymmetric electrodes). After repeated mechanical stress, I-V curves for the symmetrical structure show a significant negative threshold voltage (VT) shift, due to mechanical stress-induced oxygen vacancy generation. However, degradation in the UI structure TFTs after stress is a negative VT shift along with the parasitic transistor characteristic in the forward-operation mode, with this hump not evident in the reverse-operation mode. This asymmetrical degradation is clarified by the mechanical strain simulation of the UI TFTs. Moreover, this electrochemically induced strain can interact with the local strain existing at solid-solid interfaces. It is not clear how this interaction affects the lithiation mechanism. The effect of this coupling on the lithiation kinetics in epitaxial Fe3O4 thin film on a Nb-doped SrTiO3 substrate is investigated. In situ and ex situ transmission electron microscopy (TEM) results show that the lithiation is suppressed by the compressive interfacial strain. At the interface between the film and substrate, the existence of LixFe(3)O(4) rock-salt phase during lithiation consequently restrains the film from delamination. 2D phase-field simulation verifies the effect of strain. This work provides critical insights of understanding the solid-solid interfaces of conversion-type electrodes.Item type:Item, Cooperative Polar/Steric Strategy in Achieving Site-Selective Photocatalyzed C(sp(3))-H Functionalization(2017-06-27) ;Yamada, Keiichi ;Fukuyama, Takahide ;Fujii, Saki ;Ravelli, DavideFagnoni, MaurizioSynergistic control over the S(H)2 transition states of hydrogen abstraction exploiting polar and steric effects provides a promising cooperative strategy for site-selective C(sp(3))-H functionalization using decatungstate anion photocatalysis. By using this photocatalytic approach, the C-H bonds of substituted lactones and cyclic ketones were functionalized selectively. In the remarkable case of 2-isoamyl 4-tert-butyl cyclohexanone (1t) bearing five methyl, five methylene, and three methine C-H bonds, one methine C-H bond in the isoamyl tether was selectively functionalized.Item type:Item, Characteristics of surface plasmon coupled quantum well infrared photodetectors(2017-06-28) ;Hsu, Wei-Cheng ;Ling, Hong-Shi ;Wang, Shiang-Yu ;Lee, Chien-Ping電機學院Quantum Well Infrared Photodetectors (QWIPs) with different structures were characterized for the study of surface plasmon wave coupling. Detailed comparisons between surface plasmon coupled and etched grating coupled devices were investigated. A bias dependence for the enhancement of the responsivity of surface plasmon coupled devices was found, especially for the samples with non-uniform quantum wells. The non-uniform QWIPs with surface plasmon coupling showed an asymmetric enhancement with respect to the bias directions. Stronger enhancements were shown under the biases when a higher effective electric field region is close to the collector. The change of the photocarrier escape probability due to the narrow coupling bandwidth of the surface plasmon wave is attributed to this unexpected bias dependence. In this study, we employed annealing and hydrothermal ammonia treatments at 500 and 140 degrees C, respectively, to introduce nitrogen functionalities into GODs for enhancing their green-color PL emissions. The hydrothermal treatment preferentially produces pyridinic and amino groups, whereas the annealing treatment produces pyrrolic and amide groups. The hydrothermally treated GODs (A-GODs) present a high conjugation of the nonbonding electrons of nitrogen in pyridinic and amino groups with the aromatic pi orbital. This conjugation introduces a nitrogen nonbonding (n(N 2p)) state 0.3 eV above the oxygen nonbonding state (n(O 2p) state; the valence band maximum of the GODs). The GODs exhibit excitation-independent green-PL emissions at 530 nm with a maximum quantum yield (QY) of 12% at 470 nm excitation, whereas the A-GODs exhibit a maximum QY of 63%. The transformation of the solvent relaxation-governed pi* -> n(O 2p) transition in the GODs to the direct pi* -> n(N 2p) transition in the A-GODs possibly accounts for the substantial QY enhancement in the PL emissions. This study elucidates the role of nitrogen functionalities in the PL emissions of graphitic materials and proposes a strategy for designing the electronic structure to promote the PL performance.Item type:Item, Reaction of H plus HONO in solid para-hydrogen: infrared spectrum of (ONH)-O-center dot(OH)(2017-06-28) ;Haupa, Karolina Anna ;Tielens, Alexander Godfried Gerardus Maria ;Lee, Yuan-Pern ;應用化學系應用化學系分子科學碩博班Hydrogenation reactions in the N/O chemical network are important for an understanding of the mechanism of formation of organic molecules in dark interstellar clouds, but many reactions remain unknown. We present the results of the reaction H + HONO in solid para-hydrogen (p-H-2) at 3.3 K investigated with infrared spectra. Two methods that produced hydrogen atoms were the irradiation of HONO molecules in p-H-2 at 365 nm to produce OH radicals that reacted readily with nearby H-2 to produce mobile H atoms, and irradiation of Cl-2 molecules (co-deposited with HONO) in p-H-2 at 405 nm to produce Cl atoms that reacted, upon IR irradiation of the p-H2 matrix, readily with nearby H-2 to produce mobile H atoms. In both experiments, we assigned IR lines at 3549.6 (v(1)), 1465.0 (v(3)), 1372.2 (v(4)), 898.5/895.6 (v(6)), and 630.9 (v(7)) cm(-1) to hydroxy(oxido)-lambda(5)-azanyl radical [(ONH)-O-center dot(OH)], the primary product of HONO hydrogenation. Two weak lines at 3603.4 and 991.0 cm(-1) are tentatively assigned to the dihydroxy-lambda(5)-azanyl radical, N-center dot(OH)(2). The assignments were derived according to the consideration of possible reactions and comparison of observed vibrational wavenumbers and their IR intensities with values predicted quantum-chemically with the B3LYP/aug-cc-pVTZ method. The agreement between observed and calculated D/H- and N-15/N-14-isotopic ratios further supports these assignments. The role of this reaction in the N/O chemical network in dark interstellar clouds is discussed.Item type:Item, The dominant effect of non-centrosymmetric displacement on the crystal-field energy splitting in the strained a-plane ZnO epi-films on r-plane sapphires(2017-06-28) ;Wu, Yung-Chi ;Liu, Wei-Rein ;Chen, Hou-Ren ;Hsu, Chia-HungHsieh, Wen-FengThe polarization-dependent photoluminescence (PL) of the a-plane ZnO (a-ZnO) films grown on r-plane sapphire substrates shows that the crystal-field energy splitting (Delta(CF)) becomes larger with increasing film thickness from 24 nm to 291 nm. In the use of the nonlinear fitting of the stress-strain tensor to the X-ray diffraction data, we found crystal symmetry breaking from wurtzite to monoclinic in these films and determined the corresponding lattice constants and the relaxed lattice constants. With the available lattice variables, we further used the tight-binding method to calculate the band energies and compared with the PL spectra. The results confirm that the crystal deformation and the non-centrosymmetric displacement had opposite effects on the Delta(CF) at the Gamma point and the displacement induced by the strain is the dominant effect on Delta(CF).Item type:Item, Unveiling the hidden nematicity and spin subsystem in FeSe(2017-06-28) ;Luo, Chih-Wei ;Cheng, Po Chung ;Wang, Shun-Hung ;Chiang, Jen-CheLin, Jiunn-YuanThe nematic order (nematicity) is considered as one of the essential ingredients to understand the mechanism of Fe-based superconductivity. In most Fe-based superconductors (pnictides), nematic order is reasonably close to the antiferromagnetic order. In FeSe, in contrast, a nematic order emerges below the structure phase transition at T-s = 90 K with no magnetic order. The case of FeSe is of paramount importance to a universal picture of Fe-based superconductors. The polarized ultrafast spectroscopy provides a tool to probe simultaneously the electronic structure and the magnetic interactions through quasiparticle dynamics. Here we show that this approach reveals both the electronic and magnetic nematicity below and, surprisingly, its fluctuations far above Ts to at least 200 K. The quantitative pump-probe data clearly identify a correlation between the topology of the Fermi surface and the magnetism in all temperature regimes, thus providing profound insight into the driving factors of nematicity in FeSe and the origin of its uniqueness.
