Item type:Item, Constrained Adherable Area of Nanotopographic Surfaces Promotes Cell Migration through the Regulation of Focal Adhesion via Focal Adhesion Kinase/Racl Activation(2018-05-02) ;Lim, Jiwon ;Choi, Andrew ;Kim, Hyung Woo ;Yoon, HyungjunPark, Sang MinCell migration is crucial in physiological and pathological processes such as embryonic development and wound healing; such migration is strongly guided by the surrounding nanostructured extracellular matrix Previous studies have extensively studied the cell migration on anisotropic nanotopographic surfaces; however, only a few studies have reported cell migration on isotropic nanotopographic surfaces. We herein, for the first time, propose a novel concept of adherable area on cell migration using isotropic nanopore surfaces with sufficient nanopore depth by adopting a high aspect ratio. As the pore size of the nanopore surface was controlled to 200, 300, and 400 nm in a fixed center-to-center distance of 480 nm, it produced 86, 68, and 36% of adherable area, respectively, on the fabricated surface. A meticulous investigation of the cell migration in response to changes in the constrained adherable area of the nanotopographic surface showed 1.4-, 1.5-, and 1.6-fold increase in migration speeds and a 1.4-, 2-, and 2.5-fold decrease in the number of focal adhesions as the adherable area was decreased to 86, 68, and 36%, respectively. Furthermore, a strong activation of FAK/Racl signaling was observed to be involved in the promoted cell migration. These results suggest that the reduced adherable area promotes cell migration through decreasing the FA formation, which in turn upregulates FAK/Racl activation. The findings in this study can be utilized to control the cell migration behaviors, which is a powerful tool in the research fields involving cell migration such as promoting wound healing and tissue repair.Item type:Item, Glucosamine-Based Supramolecular Nanotubes for Human Mesenchymal Cell Therapy(2018-05-02) ;Talloj, Satish Kumar ;Cheng, Bill ;Weng, Jen-Po ;Lin, Hsin-Chieh材料科學與工程學系Herein, we demonstrate an example of glucosamine-based supramolecular hydrogels that can be used for human mesenchymal cell therapy. We designed and synthesized a series of amino acid derivatives based on a strategy of capping D-glucosamine moiety at the C-terminus and fluorinated benzyl group at the N-terminus. From a systematic study on chemical structures, we discovered that the glucosamine-based supramolecular hydrogel [pentafluorobenzyl (PFB)-F-Glu] self-assembled with one-dimensional nanotubular structures at physiological pH. The self-assembly of a newly discovered PFB-F-Glu motif is attributed to the synergistic effect of pi-pi stacking and extensive intermolecular hydrogen bonding network in aqueous medium. Notably, PFB-F-Glu nanotubes are proven to be nontoxic to human mesenchymal stem cells (hMSCs) and have been shown to enhance hMSC proliferation while maintaining their pluripotency. Retaining of pluripotency capabilities provides potentially unlimited source of undifferentiated cells for the treatment of future cell therapies. Furthermore, hMSCs cultured on PFB-F-Glu are able to secrete paracrine factors that downregulate profibrotic gene expression in lipopolysaccharide-treated human skin fibroblasts, which demonstrates that PFB-F-Glu nanotubes have the potential to be used for wound healing applications. Overall, this article addresses the importance of chemical design to generate supramolecular biomaterials for stem cell therapy.Item type:Item, Single-Crystal Antimonene Films Prepared by Molecular Beam Epitaxy: Selective Growth and Contact Resistance Reduction of the 2D Material Heterostructure(2018-05-02) ;Chen, Hsuan-An ;Sun, Hsu ;Wu, Chong-Rong ;Wang, Yu-XuanLee, Po-HsiangSingle-crystal antimonene flakes are observed on sapphire substrates after the postgrowth annealing procedure of amorphous antimony (Sb) droplets prepared by using molecular beam epitaxy at room temperature. The large wetting angles of the antimonene flakes to the sapphire substrate suggest that an alternate substrate should be adopted to obtain a continuous antimonene film. By using a bilayer MoS2/sapphire sample as the new substrate, a continuous and single-crystal antimonene film is obtained at a low growth temperature of 200 degrees C. The results are consistent with the theoretical prediction of the lower interface energy between antimonene and MoS2. The different interface energies of antimonene between sapphire and MoS2 surfaces lead to the selective growth of antimonene only atop MoS2 surfaces on a prepatterned MoS2/sapphire substrate. With similar sheet resistance to graphene, it is possible to use antimonene as the contact metal of 2D material devices. Compared with Au/Ti electrodes, a specific contact resistance reduction up to 3 orders of magnitude is observed by using the multilayer antimonene as the contact metal to MoS2. The lower contact resistance, the lower growth temperature, and the preferential growth to other 2D materials have made antimonene a promising candidate as the contact metal for 2D material devices.Item type:Item, Interaction of a pair of ferrofluid drops in a rotating magnetic field(2018-05-03) ;Qiu, Mingfeng ;Afkhami, Shahriar ;Chen, Ching-Yao ;Feng, James J.機械工程學系We use two-dimensional numerical simulation to study the interaction between a pair of ferrofluid drops suspended in a rotating uniform magnetic field. Numerical results show four distinct regimes over the range of parameters tested: independent spin, planetary motion, drop locking and direct coalescence. These are in qualitative agreement with experiments, and the transition between them can be understood from the competition between magnetophoretic forces and viscous drag. We further analyse in detail the planetary motion, i.e. the revolution of the drops around each other while each spins in phase with the external magnetic field. For drops, as opposed to solid microspheres, the interaction is dominated by viscous sweeping, a form of hydrodynamic interaction. Magnetic dipole-dipole interaction via mutual induction only plays a secondary role. This insight helps us explain novel features of the planetary revolution of the ferrofluid drops that cannot be explained by a dipole model, including the increase of the angular velocity of planetary motion with the rotational rate of the external field, and the attainment of a limit separation between the drops that is independent of the initial separation.Item type:Item, Fe Doped Magnetic Nanodiamonds Made by Ion Implantation as Contrast Agent for MRI(2018-05-04) ;Lin, Bo-Rong ;Chen, Chien-Hsu ;Kunuku, Srinivasu ;Chen, Tzung-YuangHsiao, Tung-YuanWe report in this paper a new MRI contrast agent based on magnetic nanodiamonds fabricated by Fe ion implantation. The Fe atoms that are implanted into the nanodiamonds are not in direct contact with the outside world, enabling this new contrast agent to be free from cell toxicity. The image enhancement was shown clearly through T2 weighted images. The concentration dependence of the T2 relaxation time gives a relaxivity value that is about seven times that of the regular non-magnetic nanodiamonds. Cell viability study has also been performed. It was shown that they were nearly free from cytotoxicity independent of the particle concentration used. The imaging capability demonstrated here adds a new dimension to the medical application of nanodiamonds. In the future one will be able to combine this capability of magnetic nanodiamonds with other functions through surface modifications to perform drug delivery, targeted therapy, localized thermal treatment and diagnostic imaging at the same time.Item type:Item, UV absorption spectrum of allene radical cations in solid argon(2018-05-05) ;Chin, Chih-Hao ;Lin, Meng-Yeh ;Huang, Tzu-Ping ;Wu, Yu-Jong應用化學系Electron bombardment during deposition of an Ar matrix containing a small proportion of allene generated allene cations. Further irradiation of the matrix sample at 385 nm destroyed the allene cations and formed propyne cations in solid Ar. Both cations were identified according to previously reported IR absorption bands. Using a similar technique, we recorded the ultraviolet absorption spectrum of allene cations in solid Ar. The vibrationally resolved progression recorded in the range of 266-237 nm with intervals of about 800 cm(-1) was assigned to the A E-2 <- X E-2 transition of allene cations, and the broad continuum absorption recorded in the region of 229-214 nm was assigned to their B (2)A(1) <- X E-2 transition. These assignments were made based on the observed photolytic behavior of the progressions and the vertical excitation energies and oscillator strengths calculated using time-dependent density functional theory.
