Item type:Item, Plate-based biochemical assay using quantum dots as a fluorescent labeling agent(2005-07-22) ;Huang, CP ;Liu, HW ;Tsao, CY ;Yin, LTChiu, SFA biochemical assay based on fluorescence imaging analysis by confocal laser scanning as a detection technique is specified. The method involved a glass plate coated with nitrocellulose (NC), where alpha-human-IgE-biotin at various concentrations was immobilized on an NC plate. Luminescent core-shell CdSe/ZnS QD-SA conjugates were then used as a fluorescent labeling agent to be captured specifically by biotinated alpha-human-IgE immobilized in a micro-array by affinity binding between SA and biotin in a plate-based biochemical assay. A confocal laser scanner was used to detect the fluorescence signals from the alpha-human-IgE-biotin-SA-QD complex. Experimental findings reveal that fluorescence intensity of QD-SA saturated at QD concentrations above 0.4 mg/ml. Moreover, a calibration curve between the fluorescence intensity and the concentration of alpha-human-IgE-biotin is plotted. The range of detectable concentrations of biotinlated alpha-human IgE was found to be between 3.3 and 100 mu g/ml in the a-human-IgE-biotin-SA-QD complex. Therefore, the results were consistent with the binding specificity in a plate-based biochemical assay, as determined using luminescent QDs as a labeling agent. (c) 2004 Elsevier B.V. The photoluminescence (PL) intensity of a triplet emitter [bis(4-trifluoromethyl)-2-phenylbenzothiazolatoacetylacetonate-iridium(III) (Ir complex 1) and bis(4-methyl)-2-phenylbenzothiazolatoacetylacetonate-iridium(III) (Ir-complex 2)] was dramatically enhanced when bluish-green emitting CdSe/ZnS QDs were incorporated into these compounds. Experimental results indicate that the emissive region of QDs substantially overlaps with the low-energy absorption bands of Ir-complexes, indicating that the photons were absorbed by both the QDs and the Ir-complexes and that the energy absorbed by the QDs was transferred efficiently to the Ir-complex triplet emitter, resulting in the observed enhancement of PL intensity. A slow quenching of QDs emission was observed in a prepared set of solutions with gradual increasing of Ir complex concentration at a fixed QDs concentration in thin film PMMA matrix, which supports the energy transfer from Ir complex to QDs. In the fabricated double-layer electroluminescent (EL) devices, the emitting layer contained either only Ir-complex or a mixture of Ir-complex and CdSe/ZnS QDs with a specific molar ratio [Ir-complex/QDs = 1/0 (D-I); 1/0.5 (D-II),..., 1/10 (D-V)]. The EL intensity and the luminance efficiency for D-II were higher (luminescence yield = 19.3cd A(-1)) than those of the other devices. The PL and EL enhancement of the triplet emitter were also strongly supported by using ZnSe QDs rather than CdSe/ZnS QDs.Item type:Item, Thermodynamic parameters of iron-cyanide adsorption onto gamma-Al2O3(1997-04-15) ;Huang, CP ;Cheng, WP ;環境工程研究所Institute of Environmental EngineeringIn this study, we describe the adsorption of ferricyanide (Fe(CN)(6)(3-)) on gamma-Al2O3 by a modified Langmuir adsorption isotherm. The modified Langmuir isotherm developed herein can accurately predict the apparent equilibrium constant (K-app) at different pH values and temperatures. In the experimental pH range of 5-6.5, adsorption capacity and apparent equilibrium constants decrease as the temperature increases. Because surface protonation increases as pH decrease, a more positively charged site on the surface increases the attraction force that exists between gamma-Al2O3 and the adsorbate. According to the Gibbs-Hehmholtz equation, these apparent equilibrium constants at various temperatures and pH conditions can be applied to determine enthalpy changes (Delta H-0) for the adsorption at different pH conditions. The result indicates that enthalpy change at different pH values is near -5 kcal/mol which indicates that the enthalpy change is in the order of hydrogen bonding and confirms the exothermic nature of the adsorption process. The results also indicate that other thermodynamic parameters change of ferricyanide adsorption. The positive entropy change (Delta S-0) has been explained from the viewpoint of exchange reaction in this study. The negative free energy change (Delta G(ads)(0)) for ferricyanide adsorption shows the spontaneous nature of the adsorption process. A strong correlation between alum chemistry and the dewatering efficiency exists. At pH 3, where the positively charged ions are the dominating species, the alum addition has only a minor improvement on dewatering efficiency. At pH 7 and pH 10, where sweep flocs enmeshment play the major role, dewatering performance improves continuously as alum dose increases. At pH 5.2, where a charge reversal regime exists, the dewatering efficiency exhibits a reversal phenomenon as well. The overall dewaterability follows: pH 5.2 > pH 3 > pH 7 > pH 10. A complete test consists of the filtration and expression stages, which are discussed separately. The filtration stage is discussed based on the average specific resistance and the residual moisture in the filtration cake. The expression stage is discussed based on the combined Terzaghi-Voigt rheological model, from which the fraction contributed by the secondary consolidation (B) and the particle creeping factor (eta) are evaluated. For the dewatering sake, a lower B and a higher eta are desired. Both parameters B and eta increase at elevated alum dose and/or lower pH value, while exhibiting a local minimum in charge reversal regime at pH 5.2. The optimal alum dose considering sludge dewaterability is discussed.Item type:Item, Biotreatment of ammonia from air by an immobilized Arthrobacter oxydans CH8 biofilter(1997-11-01) ;Chung, YC ;Huang, CP ;Tseng, CP ;環境工程研究所生物科技學系A heterotrophic Arthrobacter oxydans CH8 that was capable of removing NH3 from NH3 containing gas was isolated from livestock farming wastewater. The A. oxydans CH8 was immobilized with calcium alginate packed into filter column. Metered NH3-containing gas was partially humidified and passed through the glass column. Extensive tests including the removal characteristics, the removal efficiencies, and the metabolic products of NH3 by A. oxydans CH8 were conducted. Additionally, the operation criteria for the biofilter was also established. NH3 removal capacities were elevated by the immobilized-cell (biological conversion) method and the BDST (bed depth service time) method (physical adsorption), respectively. The optimum temperature for removing NH3 was 30 degrees C, while the nitrification ability remained 80% at 40 degrees C. The high efficiency (>97%) in the removal of NH3 was attained at 36 L/h with pH control and was not decreased because of high NH3 inlet concentration. In addition, the high maximum removal rate (1.22 g of N/day.(kg of bead)) enhanced the use of the biofilter in industrial-scale NH3(g) pollution control. The ability to remove NH3 at high inlet concentration and temperature suggested that the immobilized A. oxydans CH8 biofilter has potential in processing NH3 gas.Item type:Item, Interactions between alum and organics in coagulation(1996-08-01) ;Huang, CP ;Shiu, HL ;環境工程研究所Institute of Environmental EngineeringThe coagulation-sedimentation process, i.e. a major pretreatment for filtration, has the potential to remove organic contaminants. However, interactions between coagulants and organics in the absence of suspended solids have only received limited attention. This study aims to model alum-organics' interactions corresponding to the behavior of organics during coagulation. Two typical organic materials, humic acid and salicylic acid, are investigated. Experimental results indicate that the removal of organics by coagulation is directly related to the molecular weight. The interaction of alum with humic acid involves complexation, charge neutralization, precipitation and adsorption. The coagulation of salicylic acid with alum arises from adsorption and entrapment. Results of this study confirm a stoichiometric relationship between the initial concentrations of humic acid and optimal alum dosages.Item type:Item, Adsorption characteristics of iron-cyanide complex on gamma-Al2O3(1996-08-10) ;Cheng, WP ;Huang, CP ;環境工程研究所Institute of Environmental EngineeringThe adsorption behavior of ferrocyanide and ferricyanide toward aluminum oxide with a 0.01 M NaClO4 background electrolyte at 25 degrees C was investigated. Results obtained from different pH values demonstrated that the adsorption of these complex ions could be described by a modified Langmuir isotherm in the concentration range 0.5 X 10(-4) to 4 X 10(-4) M. This modified Langmuir isotherm developed here could accurately predict the equilibrium partition between solid and liquid phases while taking proton competition into account. Also, the apparent equilibrium constant (K-app) derived from the Langmuir equation for ferrocyanide or ferricyanide adsorption on aluminum oxide was compared with the intrinsic constant (K-int) calculated by triple-layer model simulation. This comparison revealed a strong correlation between the apparent equilibrium constant and the intrinsic constant. Moreover, in comparing K-app with K-int, we can infer that the adsorption of either ferrocyanide or ferricyanide complexes onto aluminum oxide is achieved through outer-sphere complexation. Prior to simulation testing, a series of batch tests was performed to determine the optimum state of dosing corresponding to the monitor's output. The results indicate that floc density decreased as floc size increased under the same coagulant dosage. Moreover, ratio output of the monitor could provide an index for evaluating the change trend of flee size in flocculation and for predicting the potential of flee settling. An automatic control system was designed and connected to a laboratory-scale water treatment process. A computer in the automatic control system received all the ratio values with respect to the progressive dosing. After determining the dosage, the system terminated the function of control. This control system started performing a new test for determining another optimum dosage when the fluctuation of inflow turbidity was more than a designated value. Simulation tests demonstrated that the PDA monitor with a designed control system could be successfully applied in the on-line control of coagulation dosing. The recommended control parameters for the control system was also identified. The saturation constants for H2S by Thiobacillus thioparus CH11 and Pseudomonas putida CH11 were calculated to be 30.3 ppm and 44.2 ppm, respectively.Item type:Item, Application of Aspergillus oryzae and Rhizopus oryzae for Cu(II) removal(1996-09-01) ;Huang, CP ;Huang, CP ;環境工程研究所Institute of Environmental EngineeringBiosorption of copper(II) by the untreated and acid-treated fungal biomass has been studied in both batch and column modes. Two species, Aspergillus oryzae and Rhizopus oryzae, were used in this study. A. oryzae mycelia (undergoing acid-washing) exhibit a clear advantage in Cu(II) removal, compared with other adsorbents. The acid-washing process can strongly enhance the adsorption capacity for A. oryzae mycelia. However, acid-washing does not alter the metal adsorption capacity of R. oryzae mycelia. The result indicates that acid-washing is not only a pretreatment step but also a regeneration step in the heavy metal removal process. These dual purposes, therefore, give the acid-washing biomass an indication of successful prospect. Cultivating A. oryzae in pellet form is an effective means of mycelium immobilization. The method established in this study provides the high-yield, uniformly-sized particles (2-3 mm in diameter), which are effective in solid-liquid separation. This pellet column can completely remove metals before breakpoint. After the breakpoint, a significant amount of Cu(II) removal over a long period has been observed. This is thought to be the result of intracellular uptake. Copyright (C) 1996 Elsevier Science Ltd
