dc.description.abstract在此揭示可自我斷裂的胜肽連接子,其可用來生產純化蛋白質。所述之可自我斷裂的胜肽連接子是插入到幾丁質結合蛋白(CBP)與一標的蛋白之間,進而形成一種融合蛋白。讓此融合蛋白通過一幾丁質基質管柱,使此融合蛋白中的CBP部分與幾丁質基質結合,接著讓融合蛋白中該可自我斷裂的胜肽連接子在pH值介於約5.5-7.5間的緩衝溶液中進行自我斷裂,以釋出標的蛋白。此幾丁質基質管柱還可以pH值介於約3-4間的緩衝溶液加以再生,亦即,將其上所結合的CBP洗出,回復成原先的幾丁質基質,此幾丁質基質可重複使用至少6次仍不會喪失活性。zh_TW
