dc.description.abstract | 硫化氫為生體內重要的小分子,在人體內很多重要的生理功能具有不可或缺的角色,如果能以磁振造影的方式偵測體內的硫化氫,對於未來在分子醫學顯影診斷中,提供磁振造影儀在臨床有更多的使用性,在本研究中,我們設計並合成一個同時具有釓金屬錯合物端及具有帶氟官能基之高水溶性化合物[Gd (DOTA-CF3)],目的是利用高順磁性金屬-釓與氟原子之間的順磁性增強弛緩效應(PRE effect)使19F MRI的訊號被遮蔽,當與生物硫醇反應後,造成氟官能基游離出來,讓19F的訊號被偵測到,實驗結果得知,此化合物[Gd (DOTA-CF3)]在生物硫醇分子存在下會反應,尤其對硫化氫更具較佳的反應性,同時以NMR偵測19F的訊號,得到訊號被遮蔽及顯露的結果,確定此化合物設計的原理對於偵測硫化氫是具有可行性的。 | zh_TW |
dc.description.abstract | H2S, an important small molecule in the body, performs an indispensable role in many biological activities. H2S detection using MRI in human body plays a crucial role in Molecular Imaging clinical trial with potential outcome. In this study, we designed a Gadolinium complex [Gd(DOTA-CF3)] containing a hydrophilic ligand with fluorine as the functional site to detect the H2S. Upon reaction with biothiols the PRE effect in complex enhances the paramagnetism between Gadolinium and fluorine and this helps to shorten 19F MRI signals which can be detected easily using 19F-NMR. Results showed that [Gd(DOTA-CF3)] selectively detects H2S among the biothiols . | en_US |
