dc.description.abstract本論文以 BODIPY 為螢光基團,合成螢光感測分子CBS。化合物 CBS 藉由綠色螢光 turn-on 的方式來辨識 Cu2+ 離子,在其他金屬離子的存在下,仍對 Cu2+ 具有選擇性。且於緩衝溶液中仍保有金屬離子選擇性,並能在 pH 5.0~9.0 範圍偵測 Cu2+ 之存在。由 Job plot 得知 CBS-Cu2+ 採 1:1 比例錯合。化合物CBS對Cu2+結合常數為7.28 × 103 M-1、偵測極限則是0.87 μM;此外,其更可進一步應用於細胞實驗,於RAW 264.7 cells內表現偵測Cu2+之能力。zh_TW
dc.description.abstractIn this thesis, we have developed a BODIPY-based fluorescent chemosensor CBS for metal ion sensing. The chemosensor CBS containing a Se2N moiety exhibited high selectivity for Cu2+ detection. Significant fluorescence enhancement was observed with chemosensor CBS in the presence of Cu2+;however, the metal ions Ag+, Ca2+, Cr3+, Co2+, Fe2+, Fe3+, Hg2+, K+, Mg2+, Mn2+, Ni2+, Pb2+ and Zn2+ produced only minor changes in fluorescence values for the system. The apparent binding constant (Ka) of Cu2+ binding in chemosensor CBS was found to be 7.28 × 103 M-1. The binding ratio of the CBS-Cu2+ complexes was determined as 1:1 according to Job plot. The maximum fluorescence enhancement caused by Cu2+ binding in chemosensor CBS was observed in the pH range of 5.0 ~ 9.0. In addition, chemosensor CBS can be used as a fluorescent probe for detecting Cu2+ in living cells.en_US
