dc.description.abstract | 近年來,應用毛細管電泳之微胞電動層析法(micellar electrokinetic chromatography, MEKC)在傳統中藥複方的指標成份分析研究上已有相當進展,且由於毛細管電泳具有較高的分離效率,分析時間短,所需樣品量少,再現性佳,操作簡單、容易自動化等優點,其在中藥材成份分析的領域已逐漸與HPLC分庭抗禮。 本篇論文分別針對柴胡桂枝湯、小柴胡湯,及真武湯、四逆湯、苓桂朮甘湯、苓薑朮甘湯、小半夏加茯苓湯等七種常被使用而藥材組成重複性大的複方湯劑進行毛細管電泳分析方法的建立,並對各分析條件進行討論。中藥複方的樣品基質相當複雜,將以適當的樣品前處理配合所建立的毛細管電泳分離條件,進行分析。 在尋找毛細管電泳分離條件方面,藉由界面活性劑與環糊精的加入,及緩衝溶液濃度的調整,已可獲得分離效率高、分析時間短的分析方法,且可應用於實際樣品的分析上。 | zh_TW |
dc.description.abstract | In this thesis, the high-performance micellar electrokinetic chromatography (MEKC) method with diode-array detection was developed to analyze specific ingredients in seven traditional Chinese medicinal preparations. These preparations include Chai-hu-kuei-chih-tang, Hsiao-chai-hu-tang, Chen-wu-tang, Szu-ni-tang, Ling-kuei-chu-kan-tang, Ling-chiang-chu-kan-tang and Hsiao-pan-hsia-chia-fu-ling-tang. The factors affecting the separation efficiencies were examined by combining different micelles and modifiers. Twelve specific components in Chai-hu-kuei-chih-tang and Hsiao-chai-hu-tang were well separated with a MEKC method. The running buffer was a pH 9.3 borate buffer containing SDS. The entire separation was completed within 13 minutes. The extraction efficiency of real samples with different extraction solution was evaluated. A cyclodextrin-modified MEKC method (CD-MEKC) was established to separate eight specific components in Chen-wu-tang, Szu-ni-tang, Ling-kuei-chu-kan-tang, Ling-chiang-chu-kan-tang and Hsiao-pan-hsia-chia-fu-ling-tang. The optimized separation buffer was a pH 9.0 borate buffer containing SDS and γ-cyclodextrin. The whole separation time was less than 11 minutes. Those two MEKC methods pose high separation efficiencies and high precision. Therefore, MEKC method could be a suitable technique for further study of traditional Chinese medicines. | en_US |
