摘要: 台灣蜆(Corbicula fluminea)為台灣主要的淡水養殖貝類,具有很高的經濟價值.傳統民間更視蜆為保肝良品,對於肝病是良好的營養補品。現今的研究報告指出,蜆之熱水萃出物可降低因酒精誘發的肝臟損傷效應.然而目前尚未有明確的報告指出蜆對發炎反應的影響,故本研究主要在探討蜆粗萃物的抗發炎活性。將蜆分別以氯仿及熱水萃取而得到蜆粗萃物,進一步觀察蜆粗萃物對發炎反應的影響。 本計畫中,首先將分析蜆氯仿萃取物和熱水萃取物影響THP-1 細胞株及人類周邊血液單核球細胞的生長情形及刺激細胞分泌促發炎激素的狀況,進一步將THP-1細胞株及人類周邊血液單核球細胞以脂多醣 (LPS)刺激後,蜆氯仿萃取物可藉由抑制促發炎激素的mRNA表現進而抑制細胞對促發炎激素的釋放,但熱水萃取物對細胞分泌促發炎激素的影響則尚不明確。隨後以in vivo的實驗探討蜆氯仿粗萃物的抗發炎活性,進一步證實蜆氯仿粗萃物的抗發炎活性和in vitro實驗結果一致。並利用GC-MS質譜儀分析蜆氯仿粗萃物中所含成分,發現蜆氯仿粗萃物中至少含有六種以上已被文獻報導具有抗發炎潛力的活性成分。藉由以上實驗,本研究展現出台灣蜆抗發炎的潛力,而蜆氯仿萃取物所含的成分在抗發炎活性中扮演重要的角色。
Corbicula fluminea(Freshwater clam)is the major fresh water cultivated bivalvia in Taiwan. It is used to be as a remedy for liver diseases such as hepatitis. Recent reports indicate that the hot water extracts of C. fluminea could reduce liver injury induced by ethyl alcohol. However, whether C. fluminea has any influence on inflammation is still unclear. For evaluation of C. fluminea’s anti-inflammatory activity, masses of the claim were respectively extracted by chloroform or hot water to obtain the clam chloroform extracts (CCE) and hot water extracts (CWE). Briefly, CCE and CWE were examined their influences on proinflammatory responses in this study. Firstly, the results revealed that neither CCE nor CWE could induce changes in proliferation of human mononuclear cells and the releases of proinflammatory cytokines. However, when the inflammatory responses were triggered by lipopolysaccharide (LPS), CCEs significantly displayed their anti-proinflammatory activities to lower the releases of LPS-induced proinflammatory cytokines through suppressing mRNA expression. Unlike CCE, CWEs showed their minor and controversial actions on the releases of LPS-induced proinflammatory cytokines. Sequentially, CCEs were further verified their anti-inflammatory activities in vivo and the results were consistent with the actions of CCEs in vitro. Finally, the CCEs were determined their components by GC-MS and the results exposed that there were at least 6 compounds in CCEs with various anti-inflammatory activities according to the published literatures. Thus, this study revealed C. fluminea has the anti-inflammatory capability and indicated that the compounds in CCE play the key roles of anti-inflammatory activity.
