摘要: | 微全分析系統(micro-total analysis system,μ-TAS)是一種微小化的整合分析元件,
結合了進樣、分離與檢測等功能在一片微晶片中,而微晶片電泳(microchip capillary
electrophoresis)即是以電泳機制將樣品在微管道中分離,並在末端檢測分析物的微分析
系統。這種微分析系統具有快速分離,高分離效率及解析度,可攜式,大量分析,以
及降低成本及廢液等特點。電化學檢測法係利用分析物本身的氧化還原特性來進行檢
測,不需複雜的前處理或衍生化反應,是一種適用性相當廣的檢測方法。本研究將發
展簡單的微流體晶片結合電化學檢測器,測試不同的電極,並配合電極修飾,使其成
為再現性佳、應用性廣的分析元件,並可針對多種化合物進行快速分析。本研究計畫
也將探討一些影響微晶片電泳分離效率的因素及相關對策,利用不同的方法加強微晶
片製作的再現性,提高微晶片電泳檢測的靈敏度,探討微晶片電泳/電激發化學放光法
整合晶片的製作及應用,同時結合毛細管電泳,研究一些重要的生化反應。 Micro-total analysis system is an integrated analytical system containing injection, separation, and detection in a miniaturized microchip. Microchip capillary electrophoresis (microchip CE) has been demonstrated by employing electrophoretic separation in a microchip integrated with various detection methods. Microchip CE possesses the advantages of the miniaturized separation systems, such as rapid analysis, high separation efficiency and resolution, portability, increased throughput, and reduced cost and waste. Electrochemical detection is ideally suited to miniaturized analytical system and is an attractive alternative detection mode for microchip CE. We will study the effects of integrating various electrodes with microchip CE to analyze different compounds. Effective electrode modification will also be used to improve the selectivity and sensitivity. In order to have a better understanding how different experimental setups will affect separation performance and signal intensity in the microchip CE, various experimental conditions will be manipulated. Both microchip CE and capillary electrophoresis methods will be used to investigate important biological compounds. |
