摘要: | 我们测试了三类(全氟、阴离子型、非离子型)共八种的界面活性剂在25bar下、液体R134a冷媒中的溶解度,发现只有二种非离子型界面活性剂Polyethylene glycol mono-4-nonylphenyl ether(PGMNPE)与Polyethylene glycol monooleyl ether(PGME)能够溶解。将PGMNPE/PGME溶于25bar的液体R134a冷媒中来萃取固体的蛋白质,初步的萤光光谱测量结果显示,似乎可将溶菌□ (lysozyme)、胰朊□(trypsin)、胃液□(pepsin)及牛血清蛋白(bovine serum albumin,BSA)四种固体蛋白质萃取出来。然而进一步的实验证实,萤光的讯号来自于冷媒R134a中的不纯物所造成的干扰。但是可见光光谱的资料显示,PGME溶于25bar液体R134a冷媒的系统确实能将三种色素:甲基橙(methyl orange)、原藻红(erythrosine)与孟加拉玫瑰素(rose bengal)萃取出来。我们希望能由溶剂极性与色素吸收光谱位移的关系来判断萃取的机制是否为形成反微胞,但是尚无法由实验结果得到证明。 We investigated the solubility of eight surfactants in coolant R134a. Among them, two non-ionic surfactants, i.e. polyethylene glycol mono-4-nonylphenyl ether (PGMNPE) and polyethylene glycol monooleyl ether (PGME) were soluble. These two compounds were then added to R134a individually in order to examine the feasibility of solubilizing proteins. According to the preliminary fluorescence data, it seemed that lysozyme, trypsin, pepsin and bovine serum albumin were all soluble in R134a at the presence of either PGMNPE or PGME. Further studies, however, validated that the supposed protein fluorescent signals actually were originated from impurities of R134a. In the course of investigating other polar compounds, we found that three colorants, i.e. methyl orange, erythrosine and rose bengal, were indeed solublized in the system. As to the mechanism accounted for this solubilizing, there still lacked solid evidence to confirm the formation of reverse micelles. |
