摘要: 近年来,高分子材料在生医科技的应用上具有举足轻重的地位。本研究计画旨在发
展新型高分子团联共聚物,以做为治疗癌症之“奈米药物载体”与组织修复之“多孔性
支架”。本计画之高分子团联共聚物是由生分解高分子PLA 与类磷脂质高分子PMPC 所
组成,兼具高生物相容性与生物分解性。在体内可避免免疫系统辨识、有效地抑制蛋白
质及血小板之吸附。因此具有良好的血液稳定性而使高分子奈米微胞滞留在体内的时间
增长,并藉由癌症组织EPR 效应(enhanced permeability and retention effect)大量累积
于癌细胞,减少对正常细胞之伤害。此外,亦可改变高分子组成比例,制备成具良好机
械强度之多孔性支架,以组织工程技术培养细胞于支架内,以做为关节软骨治疗之基
材。
In the recent years, polymeric materials play an important role in the application
of bio- and medical-technologies. The main goal of this research is to develop a novel
polymeric block copolymer for cancer therapy and tissue engineering. The novel
block copolymer of this project is composed by biodegradable polymer of PLA and
lipid-like polymer of PMPC. It owns not only high biocompatibility, but also
biodegradability and bioacceptability. In our first section, nanoparticles with
core-shell structure will be prepared by the self-assembly of copolymers. They can
avoid the recognization by MPS and the adhesion of proteins or platelets in serum.
Via the EPR effects, nanoparticles can be accumulated in tumor tissue naturally. This
mechanism will arise the efficacy of cancer therapy and reduce the cytotoxicity of
normal tissue. In addition, porous scaffold will also be prepared in our second section.
This scaffold will be a matrix for cell adhesion and growth via the technology of
tissue engineering.
