摘要: 生物分子和電子元件的結合,是21世紀人類積極突破的新科技,如何進一步將數十年來累積的電子資訊雄厚根基,連結到生命演化的智慧,且發展出生物感測、運算、記憶、微處理器等技術,是生物電子研發的目標。生物電子元件大致上可分為氣態環境與液態環境兩類。本計劃在主持人於有機電子元件以及非晶銦鎵鋅氧化物半導體薄膜電晶體(a-IGZO TFT)的研究基礎下,特別提出兩種極具潛力的新穎電子生化感測器,分別針對氣態偵測和液態環境偵測進行研究。針對氣態環境下:由於人體呼吸中不同的氣體成分分別標示著身體狀態的意義,例如:氨氣標示著肝臟及腎臟的生理狀態、糖尿病患者呼氣中的丙酮濃度較高等。本團隊發明了獨特的多孔性有機二極體作為氣體感測元件,感測的氨氣濃度能低於50ppb,且具備了低成本、製作簡單快速、常溫下操作、即時性感測等優勢。在本計劃中,規劃了不同的氣體成分的偵測器發明,以及與計畫之協同主持人--榮總神經再生中心主任鄭宏志醫師合作,進行針對不同呼氣氣體檢測的老鼠實驗,以期望未來能進一步用於非侵入性(noninvasive)的醫療診斷。另一方面,本團隊提出新穎的有機無機複合式生化感測器,利用a-IGZO TFT為基本元件,覆蓋或官能化不同的感測分子,利用TFT的基板效應或雙閘極效應來進行感測,a-IGZO TFT具有穩定、高電子遷移率,與製程簡單、便宜快速的優勢,本團隊先期成果中已經證實此新穎元件具有靈敏的氣體感測能力,本計劃將發展液態環境的感測器。液態環境下的感測器應用層面橫跨了:基因工程,癌症治療,疾病偵測,訊息傳遞動力學等研究。本計劃鎖定了液態環境之下的訊息分子:一氧化氮;蛋白質Biotin與Avidin;更進一步的人乳頭瘤病毒(HPV-DNA)和B肝病毒(HBV-DNA)試驗。希望以本團隊現有經驗為基礎,未來能在非侵入性診斷與基礎醫療研究盡一份心力。
In the 21st century, integrating biological molecules with electronic devices is a breakthrough new technology. Using the experience of electronic technology to develop new bio/chemical sensor is the goal of bio-electronic research. Application of biosensors can be divided into the gaseous sensor and aqueous sensor. In this project, based on the former results, we propose two kinds of new bio/medical sensor. One can be applied as a sensitive gas sensor for the development of non-invasive diagnosis based on the breach analysis. The other is a new hybrid sensor based on amorphous In-Ga-Zn-Oxide TFT (a-IGZO TFT) functionalized with a sensing capping layer. For the gaseous sensing, analyzing the chemical composition of human breath helps people to examine their health conditions. It is a convinent method for a non-invasive diagnosis of disease and has been used for centuries. More than 200 organic or inorganic gaseous molecules were examined in human breath. These gaseous species were produced through normal physiological processes or pathological conditions such as gastric ulcer, liver disease, cancer, or renal failure. Recently, our team invented a gas sensor with a unique vertical structure based on a porous organic diode. The sensing limit to ammonia of this vertical type sensor is lower than 50ppb. The proposed sensor has many advantages such as room temperature operation, real-time sensing, low-cost, and rapid/simple production. In this project, we plan to develop the gas sensors targeting on different gas composition. With the support from the co-PI of this project, Dr. Prof. Henrich Cheng in Veterans General Hospital-Taipei, animal experiments with different target gases are proceeded. For aqueous sensing, we plan to develop a-IGZO TFT sensor in aqueous environment based on its high stability, high electron mobility, simple and inexpensive production cost. In aqueous environment, sensor applications include: genetic engineering, cancer treatment, disease detection, signaling dynamics study. In this part of the plan, we are concerned in signaling molecule - "Nitric oxide", protein (Biotin and Avidin pair), and further human papillomavirus (HPV-DNA) and hepatitis B virus (HBV-DNA) test. Based on our experience and the support from the co-PIs, we believe that we can make a contribution to non-invasive diagnosis and basic medical research.
